PCR与膜芯片分型法检测女性生殖道HPV结果比较分析 被引量:3

摘要 目的分析荧光探针PCR与膜芯片分型技术两种方法检测女性生殖道人乳头状瘤病毒(HPV)的结果,对两种方法进行临床应用评价。方法随机收取2015年01月~11月来本院门诊就诊的女性患者1062例,利用荧光探针PCR定性检测14种常见的高危型HPV病毒(包括HPV16,18,31,33,35,39,45,51,52,56,58,59,68,66),并对其中的HPV16和HPV18型进行分型检测,同时用膜芯片分型技术检测23种HPV型,包括上述14种常见高危HPV外,还包括4种高危型(HPV53,73,82,83)和5种低危型HPV病毒(HPV6,11,42,43,81),并对两种方法检测的结果进行对比分析。结果荧光探针PCR定性检测HPV的阳性率为12.90%(137/1062),膜芯片分型技术检测HPV阳性率19.68%(209/1062),阳性率明显高于探针PCR法,两者阳性率之间差异有统计学意义(χ~2=15.627,P<0.05);但两种方法检测14种常见的高危型HPV(HPV16,18,31,33,35,39,45,51,52,56,58,59,68,66)病毒阳性率之间差异无统计学意义(χ~2=1.063,P>0.05),总符合率为91.15%(968/1062),阳性符合率为91.97%(126/137),阴性符合率为91.03%(842/925),两者检测结果具有较好的一致性。结论荧光探针PCR与膜芯片分型技术检测常见的14种高危型HPV的结果具有良好的一致性,阳性率之间差异无统计学意义,但膜芯片分型技术检测HPV的基因型数比荧光探针PCR要多,检出HPV阳性率要高,更适合女性大规模健康体检。 objective:To analyze the fluorescence probe PCR chip parting with membrane technology in two kinds of methods to detect the female reproductive theater papilloma virus(HPV)as a result,the two methods for clinical evaluation. Method:Random charge from January to November 2015 to our hospital clinic on female patient 1062 examples,using the fluorescent probe PCR qualitative detection of 14 kinds of common high-risk type HPV(including HPV16,18,31,33,35,39,45,51,52,56,58,59,68,66),and the type of HPV16 and HPV18 classification tests,at the same time with membrane chip classification technology 23 types of HPV detection,including the above 14 kinds of common high-risk HPV,include four high-risk type(HPV53,73,82,73)and five kinds of low-risk type HPV(HPV6,11,42,43,81),and the testing results of the two methods were analyzed. Results:Fluorescent probe PCR qualitative detection of HPV positive rate was 12.90%(137/1062),membrane chip classification technology to detect HPV positive rate 19.68%(209/1062),were significantly higher than the probe PCR method,the difference was statistically significant between two positive rate(χ^2=15.627,P0.05);But two methods of detecting 14 kinds of common high-risk type HPV(HPV16,18,31,33,35,39,45,51,52,56,58,59,68,66)there was no statistically significant difference between virus positive rate(χ^2=1.063,P 0.05),the total coincidence rate was 91.15%(968/1062),the positive coincidence rate was 91.97%(126/137),negative coincidence rate was 91.03%(842/925),the test results have good consistency. Conclusion:fluorescent probe PCR chip parting with membrane technology to detect common 14 kinds of high-risk type HPV results have good consistency,there was no statistically significant difference between positive rate,but membrane chip technology to detect HPV genotypes number more than fluorescent probe PCR,check out the HPV positive rate is higher,the large-scale physical examination is more suitable for women.
