摘要
由富G序列构成的G四链体(G4),作为一种非常规的DNA立体结构具有特殊的物理化学性质.本文从G四链体的特点出发,分别从多元的作用活性、结构和构象多样性以及多种信号表达方式3个角度,分析了G四链体在生物传感器中的应用潜力,并将G四链体在生物传感器构建中的信号产生策略分为直接作用、封闭/解封闭、分段G四链体探针、聚合酶辅助信号放大和富集法5种,分别例举了相关的最新工作.
G-quadruplex (G4) is formed from guanine-rich oligonucIeotides with special physicochemical proper- ties, based on which, we analyzed G4's great applicable potential for the construction of biosensors from the aspects of vibrant binding activities, different structural topologies and multiple signal output modes. The strategies of emplo- ying G4 in biosensors included: direct interaction, block/liberation, split G4 probes, polymerase-assisted amplifying strategy and enrichment on the sensor surface. Each strategy was illustrated with latest representative works.
