摘要 骨桥蛋白(Osteopontin,OPN)是一种与免疫性疾病相关的多功能蛋白,是由被激活的巨噬细胞和淋巴细胞分泌的促炎细胞因子,在多种细胞中表达并通过与2种细胞黏附分子[整合素(α9β1、ανβ3、ανβ1、α4β1)和CD44]的结合而影响、调节许多生物学功能:包括免疫应答、组织重塑和血管增生。研究发现骨桥蛋白在炎症、癌症、细胞免疫和神经退行性疾病中明显升高。本文对骨桥蛋白在免疫相关性疾病中作用的分子机制作一概述,进而为OPN介导的相关疾病的治疗提供新的思路。 Osteopontin(OPN) is a multifunctional protein associated with immune diseases and is a proinflammatory cytokine secreted by activated macrophages and lymphocytes. OPN is expressed in a variety of cells and affects the regulation of many biological functions(immune response, tissue remodeling and vascularization hyperplasia) by binding to two kinds of cell adhesion molecules: integrins(α9β1, ανβ3, ανβ1, α4β1) and CD44.Studies have shown that osteopontin is significantly elevated in inflammation, cancer, cellular immunity, and neurodegenerative diseases. This paper summarizes the molecular mechanisms underlying the role of osteopontin in immune-related diseases, and then provide new ideas for the treatment of OPN-mediated diseases.
