不同产地马兜铃药材中马兜铃总酸的含量 被引量:11

Content of total aristolochic acid in Aristolochia debilis from different localities

摘要 目的测定不同产地马兜铃药材中马兜铃总酸的含量,为马兜铃总酸限量标准的制定提供必要的数据支持。方法以马兜铃总酸含量为指标,采用L9(34)正交试验方法考察提取溶剂、溶剂用量和提取方法 3个因素,对提取条件进行优化,用紫外分光光度法检测马兜铃总酸的含量,检测波长为319 nm。结果马兜铃总酸最优的提取条件为用含1%甲酸的甲醇,料液比1∶40,超声提取30 min,提取2次;马兜铃酸Ⅰ浓度在0.005~0.030 mg·ml-1与吸光度呈良好的线性关系,A=38.046C+0.0109,r=0.9999,平均回收率为97.79%,RSD=2.09%;不同产地马兜铃种子的马兜铃总酸含量为0.357%~0.597%,果皮的马兜铃总酸含量为0.170%~0.258%。结论不同产地马兜铃药材中马兜铃总酸含量存在明显的差异,并且种子中马兜铃总酸含量明显高于果皮。 Objective The content of total aristolochic acid from different localities was determined to provide essential data for the establishment of limit standard. Methods With the content of total aristolochic acid as index,the optimum extraction condition was confirmed by investigating the factors such as solvent,solvent volume and extraction method via L9( 34) orthogonal test.The content of total aristolochic acid was determined by ultraviolet spectrophotometry at a wave length of 319 nm. Results The optimum extraction condition for total aristolochic acid was extracting twice under ultrasound for 30 min with methanol contained0. 1% formic acid in the material-to-liquid ratio 1∶ 40. The linearity of concentration of total aristolochic acid Ⅰ and absorbance was better within the range of 0. 005 - 0. 030 mg·ml-1,and the linear regression equation was A = 38. 046 C + 0. 0109( r= 0. 9999). The average recovery was 97. 79%,RSD = 2. 09%. The content of total aristolochic acid varied from 0. 357% to0. 597% in seeds,and 0. 170% to 0. 258% in rinds. Conclusion There were obvious differences in the content of total aristolochic acid from different localities,and the content of total aristolochic acid in seeds was significantly higher than that in rinds.
