摘要
目的建立芪丹复感颗粒中藁本内酯的HPLC含量测定方法。方法采用岛津LC-20AT高效液相色谱仪,并对此方法的线性范围、重复性、稳定性、加样回收率等进行考察,建立川芎中藁本内酯的含量测定方法。结果采用Agilent Eclipse XDB-C18色谱柱(4.6mm×250 mm,5μm),流动相甲醇-水(65∶35),流速1 ml·min-1,检测波长320 nm,柱温30℃。回归方程为:Y=471534+991.6(r=0.9994,0.1595~0.9571μg),平均加样回收率为97.4%(RSD=1.14%,n=6)。结论该方法简便准确,重现性好,分离度好,可用于藁本内酯的含量测定。
Objective To develop an HPLC method for determining the content of ligustilide from Qidan Fugan Granules. Methods The inear ranges,reproducibility,stabilization and recovery rate were evaluated bsaed on a SHIMADZU HPLC system to estabish the method of determining the content of ligustilide in Ligusticum Wallichii. Results The column was Agilent Eclipse XDB-C18( 4. 6mm × 250 mm,5μm); the mobile phase consisted of methanol-water( 65∶ 35); the flow rate was 1. 0ml·min-1and the UV detector was set at 320 nm under the temperature of 30℃. The regression equation of ligustilide was Y = 471534 + 991. 6( r= 0. 9994,0. 1595 - 0. 9571μg); the average recovery of ligustilide was 97. 4%( RSD = 1. 14%,n = 6). Conclusion This method is simple,accurate,replicate and suitable for the determination of ligustilide in Qidan Fugan Granules.
