摘要
目的探讨高浓度葡萄糖对人肾脏近曲小管上皮(HK-2)细胞的糖毒性及α-亚麻酸/亚油酸(ALA/LA)的改善作用。方法常规方法培养接种HK-2细胞,设置11个葡萄糖浓度,MTT法检测葡萄糖的糖毒性;设立正常对照组、高糖模型组、阳性对照组、ALA组、LA组和ALA/LA组,检测a-ALA/LA的干预作用;葡萄糖氧化酶-过氧化物酶(GOD-POD)法检测HK-2细胞的葡萄糖吸收。结果高浓度葡萄糖(≥25.3 mmol/L,培养48h)显著抑制HK-2细胞的存活率(P<0.05);培养48 h,葡萄糖浓度25.3~56.5 mmol/L可作为HK-2细胞葡萄糖毒性造模条件;适宜剂量的ALA、LA及适宜比例的ALA/LA,能够降低HK-2细胞的葡萄糖毒性,增加HK-2细胞的葡萄糖吸收。结论必需脂肪酸ALA/LA能降低葡萄糖对HK-2细胞的糖毒性,增加其葡萄糖吸收功能。
Objectives To explore the glucose toxicity on HK-2 cells and the protective effect of essential fatty acids (a-linolenic acids, ALA/linoleic acid, LA).Methods The glucose toxicity on HK-2 cells was determined by MTT assay. HK-2 cells were divided into 6 groups: control group (N), high glucose model group (G), EPA group, ALA group (A), LA group (L) and ALA/LA group (H). The glucose absorption of HK-2 cells was determined by GOD-POD assay.Results High glucose concentration (≥25.3 mmol/L,48h culture) significantly decreased survival rates of HK-2 cells (P〈0.05). The modeling conditions of glucose toxicity on HK-2 cells were glucose concentration from 25.3 mmol/L to 56.6 mmol/L and 48h culture. Suitable dose and ratio of ALA/LA could lower glucose toxicity and increase the ability of HK-2 cells to absorb glucose (P〈0.05).Conclusion ALA/LA could reduce glucose toxicity, and also increase glucose absorption of HK-2 cells.
