二冬膏对LPS致急性肺损伤大鼠肺组织TNF-α、IL-6及AQP-1、AQP-5的影响 被引量:10

Effect of Erdong Gao on TNF-α,IL-6 and AQP-1,AQP-5 of lung tissue in rats with acute lung injury induced by lipopolysaccharide

摘要 目的:研究二冬膏对LPS致急性肺损伤(ALI)模型鼠肿瘤坏死因子-α(TNF-α)、白介素6(IL-6)含量及水通道蛋白-1(AQP-1)、AQP-5蛋白表达的影响。方法:将清洁级SD大鼠随机分为6组:正常组、模型组、二冬膏10 g/kg组、二冬膏5 g/kg组、二冬膏2.5 g/kg组、地塞米松组。除正常组外,其余各组采用一次性尾静脉iv LPS 6 mg/kg复制急性肺损伤大鼠模型。首次给药在造模后0.5 h,二冬膏分别ig二冬膏生药10、5、2.5 g/kg,正常对照组与模型组ig生理盐水l ml/100g,地塞米松组腹腔ip地塞米松0.01g/kg,1次/d,连续4 d。末次给药后2 h,处死大鼠,取肺组织检测。计算肺系数、肺湿干比(W/D);采用ELISA检测大鼠肺组织TNF-α、IL-6的含量;Real-time PCR检测大鼠肺组织AQP-1、AQP-5 mRNA表达;Western bloting测定大鼠肺组织AQP-1、AQP-5蛋白的表达。结果:与正常组比较,模型组大鼠肺系数、肺湿干比(W/D)和肺组织TNF-α、IL-6含量明显升高,肺组织AQP-1、AQP-5 mRNA和AQP-1、AQP-5蛋白表达显著降低。与模型组比较,二冬膏10、5、2.5 g/kg组均能明显降低肺系数、肺湿干比(W/D)和TNF-α、IL-6含量,显著升高肺组织AQP-1、AQP-5 mRNA和AQP-1、AQP-5蛋白的表达。结论:二冬膏通过降低急性肺损伤模型鼠促炎因子TNF-α、IL-6含量,提高肺组织AQP-1、AQP-5蛋白的表达,减轻肺水肿状态,从而有效防治急性肺损伤。 Objective: To research the effect of Erdong Gao on TNF-α, IL-6 and AQP-1, AQP-5 lung tissue in rats with acute lung injury(ALI)in- duced by LPS. Method: The SD rats were randomly divided six groups:the normal group( A), the model group (B), the Erdong Gao( 10 g/ kg) group ( C ), the Erdong Gao ( 5 g/kg) group (D) , the Erdong Gao ( 2.5 g/kg) group (E) and the Dexamethasone group ( F), ten rats in each group. In addition to rats of the normal group, rats of the other groups were disposably iv LPS 6mg/kg through tail vein. The first ad- minstration was 0.5 hour after modelled, the rats in C, D, E groups were respectively given Erdong Gao (10, 5, 2.5 g/Kg), the rats in A and B groups were given 1 ml/100g normal saline, the rats in F group were iv 0.01 g/kg dexamethasone for 4 days, once a day, 2 hours after the last adminstration,the rats were sacrificed and lung tissue was tested. The lung coefficient and lung wet/dry ratio (W/D) were calculated. The contents of TNF-α,IL-6 in lung tissue were detected by ELASA method. The expressions of AQP-1 ,AQP-5 mRNA in lung tissue were tested by Real-time PCR. The expressions of AQP-1 ,AQP-5 in lung tissue were determined by Western bloting method. Result: Com- pared with A group, the lung coefficient and lung W/D and the contents of TNF-α,IL-6 in lung tissue were obviously increased( P 〈 0.01 ), the expressions of AQP-1 ,AQP-5 mRNA and AQP-1, AQP-5 in lung tissue was significantly decreased(P 〈 0.01 ) in rats of B group. Compared with B group ,the Erdong Gao( 10,5,2.5 g/kg)groups could significantly reduce the lung coefficient and lung W/D and the content of TNF-α,IL-6 in lung tissue( P 〈0.05 or P 〈0.01 ) ,which increase the expressions of AQP-I ,AQP-5 mRNA and AQP-1 ,AQP-5 in lung tissue( P 〈 0.05 or P 〈 0.01 ). Conclusions: Erdong Gao could reduce the contents of pro-inflammatory factors TNF-α,IL-6 and increase the expressions of AQP-1 ,AQP-5 in lung tissue, alleviate the state of pulmonary edema, which thus effectively preventing and treating ALI.
