摘要
目的:探讨丹参酮IIA和振动运动对2型糖尿病大鼠肝脏损伤的保护作用及其机制。方法:腹腔注射链脲佐菌素和高脂饮食相结合的方法建立大鼠2型糖尿病模型。实验分为正常组、模型组、丹参酮IIA(10 mg/kg)组、丹参酮IIA(20 mg/kg)组、振动运动组、丹参酮IIA(10 mg/kg)+振动运动组、丹参酮IIA(20 mg/kg)+振动运动组,尾静脉采血测定大鼠血糖水平,ELESA法测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性和超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量;HE染色观察肝组织细胞结构变化;Western blot法测定肝组织核因子κB(NF-κB)p65蛋白表达水平。结果:模型组空腹血糖与正常组相比显著增加,血清ALT、AST活性增加,TNF-α、IL-6和MDA含量均增加,SOD活力降低,NF-κBp65表达明显增加。丹参酮IIA(20 mg/kg)+振动运动组空腹血糖水平与模型组相比显著降低,血清ALT和AST活性降低,TNF-α、IL-6和MDA含量均降低,SOD活力升高,而NF-κBp65在肝组织核蛋白中的表达降低。结论:丹参酮IIA联合振动运动对糖尿病肝损伤具有一定的保护作用,这种保护作用与抑制氧化应激和炎症反应有关。
Objective: To study the protective effect of Tanshinone IIA combined with vibration exercise on liver injury induced by type 2 diabetes. Methods: Type 2 diabetes was induced by high-fat diet and low-dose streptozotocin. SD rats were randomly divided into: Control group; The model group; Tanshinone IIA( 10 mg/kg) ; Tanshinone IIA(20 mg/kg) ; Vibration exercise; Tanshinone IIA( 10 mg/kg) + vibration exercise; Tanshinone IIA(20 mg/kg) + vibration exercise. Blood glucose was determined by glucometer; The content of ALT, AST, TNF-α, IL45, SOD and MDA were determined by ELISA; Liver pathological changes were observed by HE staining; NF-κBp65 was measured by western blot. Results: Compared with the control group, blood glucose increased, the content of ALT, AST, TNF-α, IL-6, and MDA in- creased,the activity of SOD decreased, the expression of NF-κBp65 increased in model group. Compared with model group, blood glucose, the content of ALT, AST, TNF-α, IL-6 and MDA decreased,the activity of SOD increased, and the expression of NF-κBp65 decreased in Tanshinone IIA + vibration exercise group. Conclusion: Tanshinone IIA (20 mg/kg)combined with vibration exercise has a protective effect on liver injury induced by type 2 diabetes, which is partially via attenuating oxidative stress and inflammatory response.
