摘要
目的:观察火把花根片抑制溶血素抗体的生成作用,考察量效关系与时效关系,为确定药物的最佳给药剂量和给药时间提供参考。方法:通过给小鼠腹腔注射鸡红细胞产生溶血素抗体的模型,观察不同给药剂量和不同给药时间对小鼠溶血素抗体生成的影响,同时进行了火把花根片对首次免疫和二次免疫后溶血素抗体生成抑制作用的比较研究。结果:量效关系实验结果表明:5g生药/kg、10g生药/kg及15g生药/kg对溶血素抗体生成的抑制率分别为0%、43.6%及71.8%。时效关系实验结果表明:火把花根片15g生药/kg给药3天、5天、7天的抑制率分别为24.0%、41.7%和61.1%;给药7天后停药,第7天、第14天、第21天及第28天的抑制率分别为27.3%、34.8%、41.4%及58.8%。首次和二次免疫实验结果表明:15g生药/kg连续给药14天,第7天和第14天(二次免疫)的抑制率分别为61.1%及6.3%。结论:火把花根片对小鼠溶血素抗体的生成有明显的抑制作用,半数有效量为:9.69g生药/kg,最佳给药剂量是15g生药/kg,最佳给药时间是7天,给药7天后的维持时间不低于4周。15g生药/kg连续给药14天对第二次免疫后溶血素抗体的生成没有明显的抑制作用。该研究为火把花根片的质量控制及该药物在临床更合理的使用提供有益的试验依据。
Objective: To evaluate the inhibition efficacy of hemolysin antibody of Huobahua root tablets on both time and dose levels. And determine the optimal dose and delivery period. Methods: All mice were immunized by intraperitoneal injection of chicken red blood cells. Then different doses and different drug administration periods were taken to each mice group. The content of hemolysin antibody in blood was analyzed according to the set time point. The inhibition effects of hemolysin antibody by first and secondary immunizations were compared. Resuits: Experimental results of dosage and effect relationship showed the rates of inhibition of hemolysin antibody by different dose(5g/kg, 10g/kg, 15g/kg) were 0 % ,43.6 % and 71.8 % respectively. Experimental results of delivery period and effect relationship showed the rates of inhibition of hemolysin antibody with a dose of 15g/kg by different drug administration period(3d,5d,17d) were 24.0 % ,41.7 % and 6l. 1% respectively at Day7. After a 7-day period administration at 15g/kg, the inhibition rates of hemolysin antibody were 27.3 % ,34.8 %, 41.4 % and 58.8 % respectively at Day7,Day14,Day21 and Day28. After continuous administration for 14 days at 15g/kg, the inhibition rates of hemolysin antibody by first and secondary immunization were 61.1% and 6.3 % respectively at Day7 and Dayl4( secondary immunization). Conclusions: With increasing dose, the inhibition rate of hemolysin antibody was significantly increased. The EDs0 of Huobahua root tables is 9.69 g crude drug/kg. The optimal dosage is 15g crude drug/kg, and the best delivery time is 7 days. The action of inhibition of hemolysin antibody lasted for 28 days after a administration period of 7 days. Administration at the dose of 15g crude drug/kg for 14 days had no obvious inhibitory effect on hemolysin antibody after the secondary immunization.
