摘要
以开环聚合法制备一系列CLA低聚物;红外光谱法、偏光显微镜等方法鉴定产物结构与液晶态,评价液晶态CLA对小鼠胚胎成纤维细胞黏附和增殖的影响;以α-平滑肌肌动蛋白为指标,采用蛋白质印迹法评价CLA对成纤维细胞向肌成纤维细胞表型转化的影响。研究发现三种CLA低聚物在37℃下均处于液晶态,能够显著促进细胞黏附、增殖和α-平滑肌肌动蛋白表达。结果表明具有仿ECM结构的液晶态CLA低聚物能显著提高NIH-3T3的黏附和增殖,并对成纤维细胞表型转化具有促进作用。
A series of CLAs were synthetised by ring- opening polymerization of lactide. Their structure and liquid crystalline state were identified by infrared spectroscopy and polarizing microscope. Their effects on 3T3 adhesion and proliferation were analysed by CCK- 8. Their effects on 3T3 differentiation were analysed by western blot using α-smooth muscle actin as the indication. The results indicated that CLAs with grafting ratios of 1∶1. 5,1∶2. 5 and 1∶3. 5were in liquid crystal phase at 37 ℃. CLA can improve 3T3 adhesion proliferation and the express of α- smooth muscle actin compared to control and PLA. It was found that CLAs can self- assemble into liquid crystal and promote improved3T3 growth and differentiation.
