重组人促红细胞生成素对心梗大鼠IL-6与IL-8表达的影响 被引量:4

The effect of recombinant human erythropoietin on the expressions of IL-6 and IL-8 in myocardial infarction rats

摘要 目的探讨重组人促红细胞生成素(rHu EPO)对心梗大鼠心肌白介素-6(IL-6)与白介素-8(IL-8)表达的影响。方法 45只SD大鼠,随机分为假手术组、心肌梗死组和r HuEPO治疗组。用western blot方法检测各组大鼠梗死区周围心肌中IL-6与IL-8的表达,RT-PCR方法检测梗死区周围心肌IL-6 mRNA与IL-8 mRNA表达变化。结果心肌梗死组大鼠心肌梗死区周围心肌IL-6与IL-8蛋白与mRNA的表达水平显著高于假手术组,与心肌梗死组相比,rHuEPO治疗组心肌梗死区周围心肌IL-6与IL-8蛋白与mRNA的表达则明显降低(P<0.01)。结论 rHuEPO下调心梗大鼠心肌梗死区周围心肌IL-6与IL-8的表达。 Objective To study the effect of recombinant human erythropoietin(rHuEPO) on the expressions of IL-6 and IL-8 in myocardial infarction rats. Methods 45 SD rats were randomly divided into sham group, myocardial infarction group(MI group) and recombinant human erythropoietin group(rHuEPO group). The expressions of IL-6 and IL-8 around infarction area were examined by Western blot, and IL-6 mRNA and IL-8 mRNA around infarction area were observed by RT-PCR method. Result The expression levels of IL-6 and IL-8 protein and mRNA around infarction area were increased significantly in MI group than in sham group, but decreased significantly in rHuEPO group than in MI group, P〈0.01. Conclusion rHuEPO could down-regulate the expression levels of IL-6 and IL-8 around infarction area in myocardial infarction rats.
