摘要
目的:探讨腺苷A2A受体对新生小鼠离体延髓脑片基本节律性呼吸放电(RRDA)的调节作用。方法:制备新生小鼠离体延髓脑片,分别使用含0.0、2.5、5.0、10.0、20.0 nmol/L腺苷A2A受体激动剂CGS21680或拮抗剂SCH58261的人工脑脊液(ACSF)灌流脑片,每个浓度灌流10 min,在每个浓度灌流第6 min时,使用吸附电极记录脑片RRDA,分析吸气时程(TI)、放电积分幅度(IA)和呼吸周期(RC)。结果:CGS21680可以缩短RC,增加TI和IA(P<0.001),对RRDA有兴奋作用。SCH58261则可以延长RC,减小TI和IA(P<0.001),对RRDA有抑制作用。结论:腺苷A2A受体参与调节新生小鼠延髓RRDA,激活腺苷A2A受体对RRDA有兴奋作用。
Aim: To investigate the effect of adenosine A2A receptor on basic rhythmic respiratory discharge activity (RRDA) of medulla oblongata slice from neonatal mice. Methods: Medulla oblongata slices in vitro from neonatal mice were prepared, and then perfused with artificial cerebrospinal fluid(ACSF) which containing 0.0, 2.5, 5.0, 10.0, 20.0 nmol/L adenosine A2A receptor agonist CGS21680 or antagonist SCH58261 for 10 min. At the 6th min of perfusion, ad- sorption electrode was used to record the inhale duration (TI) , discharge integral amplitude (IA) and respiratory cycle (RC). Results : CGS21680 could shorten RC, increase TI and IA(P 〈 0. 001 ) , excite RRDA ; SCH58261 could extend RC, reduce TI and IA(P 〈0. 001 ) ,and has inhibitory effect on RRDA. Conclusion: The adenosine A2A receptor may play an excitatory role in RRDA of medulla oblongata in neonatal mice.
