济宁青山羊微卫星标记多态性分析 被引量:7

摘要 选择位于不同染色体上、具有较高多态性的12个微卫星标记,对济宁青山羊群体的遗传多样性进行研究。结果表明,CSRD247等12个微卫星标记共检测到97个等位基因,每个标记都检测到5个以上的等位基因,平均为8.08个,有效等位基因数为3.1957~6.2113个,基因频率最高的是CSRD247标记的235 bp片段(0.4583);12个微卫星标记多态信息含量(PIC)都在0.630以上,均为高度多态标记,遗传多样性丰富,其中SRCRSP5标记的多态信息含量(PIC)最高,为0.828,各标记的观测杂合度在0.4286~0.9048之间,期望杂合度在0.5981~0.8397之间,平均期望杂合度为0.7574,属于高度杂合标记和高度杂合品种,遗传变异大。本研究可为济宁青山羊品种的选种、选育及保种工作奠定基础。 Twelve microsatellite markers with high polymorphism which located on different chromosomes were selected for studying the genetic diversity of Jining grey goat. The results showed that 97 alleles were found with the 12 microsatellite markers,and each marker had more than 5 alleles( average 8. 08 alleles).The effective number of alleles was 3. 1957 ~ 6. 2113. The gene frequency of a 235 bp allele of CSRD247 marker was the highest as 0. 4583. The PIC of the 12 markers were all more than 0. 630 which belonged to highly polymorphic markers,and the genetic diversity was abundant. Among which,the highest PIC was0. 828 of SRCRSP5. The Ho of the 12 markers were 0. 4286 ~ 0. 9048 and the He were 0. 5981 ~ 0. 8397.The average He was 0. 7574. It indicated that the markers were highly heterozygous,and the Jining grey goat was a highly heterozygous breed with great genetic variation. This study provided bases for selection breeding and breed conservation of Jining grey goat.
