水合淫羊藿素对THP-1细胞增殖和凋亡的影响及机制研究

Effect of Icaritin on Proliferation and Apoptosis of THP-1 Cell and Its Mechanism

目的:探讨水合淫羊藿素(icaritin)对THP-1细胞增殖和凋亡的影响及其可能的作用机制。方法:用MTS法、流式细胞术和Hoechst 33258荧光染色检测icaritin对TP-1细胞的增殖抑制及促凋亡作用,Western blot技术检测BCL-2、BAX和Caspase-3蛋白的表达。结果:4-32μmol/L icaritin作用24-72 h对THP-1细胞有增殖抑制作用(P<0.05),且呈时间-剂量依赖关系(r=0.946);4-32μmol/L icaritin作用24和48 h,对THP-1细胞有凋亡诱导作用(P<0.05),也显示时间-剂量依赖关系(r=0.924)。icaritin作用THP-1细胞48 h后,THP-1细胞BCL-2蛋白的表达较对照组显著降低(P<0.05),而BAX和Caspase-3表达较对照组显著增高(P<0.05)。结论:Icari血对THP-1细胞有增殖抑制及促凋亡作用,icaritin可能通过线粒体途径诱导THP-1细胞的凋亡。

Objective：To investigate the effect of icaritin on the proliferation and apoptosis of THP-1 cells and its mechanism.Methods：After treated with various concentrations of icaritin,cell proliferation was detected by MTS method,and apoptosis was measured with flow cytometry and Hoechst 33258 staining.Expression of BCL-2,BAX and Caspase-3 protein in THP-1 cell was detected by Western blot.Results：After treatment with various concentrations（4-32 μmol/L） of icaritin for 24,48,72 h,the inhibition rate of cell growth significantly increased（P 〈0.05） in timedose dependent manner（r = 0.946）;and the apoptotic rate of cells significantly increased（P〈 0.05） in time-dose dependent manner（r = 0.924）.The expression of BCL-2 protein at 48 h decreased significantly in icaritin-treated group,compared with that in control group（P 〈0.05）,while the expression of BAX and Caspase3 protein at 48 h increased significantly in icaritin-treated group,compared with that in control group（P〈 0.05）.Conclusion：Icaritin can inhibit proliferation and induce apoptosis of THP-1 in vitro,Icaritin may induce apoptosis in THP-1 cells through the mitochondrial pathway.
