高效液相色谱法测定不同种植方法及采摘日期的桔梗防风龙胆黄芩和北苍术的含量 被引量:2

摘要 目的测定不同种植方法及采摘日期的桔梗、防风、龙胆、黄芩和北苍术的含量。方法采用高效液相色谱法(HPLC)法,色谱柱为Ufavour(250 mm×4.6 mm,5μm)。检测桔梗的流动相为甲醇-乙腈-0.2%磷酸水溶液(3∶25∶72),波长为204 nm;检测防风的流动相为甲醇-水(40∶60),波长为254 nm;检测龙胆的流动相为甲醇-水(25∶75);波长为270 nm;检测黄芩的流动相为甲醇-水-磷酸(47∶53∶0.2),波长为280 mn;检测北苍术的流动相为甲醇-水(79∶21),波长为340 mn。结果采摘于2015年8月27日的人工种植桔梗含量最高,其中桔梗皂苷D为0.22%;采摘于2015年10月7日的人工2年生防风1号含量最高,其中升麻素苷为0.43%,5-O-甲基维斯阿米醇苷为0.92%;人工种植的龙胆含量最高,其中龙胆苦苷为4.96%;采摘于2015年8月27日的2年生黄芩地下部分含量最高,其中黄芩苷为14.99%;苍术中苍术素含量为0.23%,低于《中国药典》标准。结论本研究采用的方法操作简便,测定结果准确可靠,为5种中药材的规范化种植及采摘奠定了基础。 Objective To determine the content of Platycodon grandiflorum, Saposhnicovia divaricata, Radix gentianae, Radix scutellariae and Atractylodes chinensis of different planting methods and harvest dates. Methods The HPLC method was used, and the chromatographic column was Ufavour(250 mm ×4.6 mm, 5 μm). The mobile phase of Platycodon grandiflorum was methanol-acetonitrile-0.2% phosphoric acid solution(3∶25∶72), and the wavelength was 204 nm. The mobile phase of Saposhnicovia divaricata was methanol-water(40∶60), and the wavelength was254 nm. The mobile phase of Radix gentianae was methanol-water(25∶75), and the wavelength was 270 nm. The mobile phase of Radix scutellariae was methanol-water-phosphoric acid(47∶53∶0.2), and the wavelength was 280 mn. The mobile phase of Atractylodes chinensis was methanol-water(79 ∶21), and the wavelength was 340 mn. Results The content of artificial cultivated Platycodon grandiflorum harvested on 27 th August 2015 was the highest, including0.22% Platycodin D. The content of artificial cultivated 2-year-old Saposhnicovia divaricata No.1 harvested on 7th October 2015 was the highest, including 0.43% Cimicifugin and 0.92% 5-O-methylvisamidoside. The content of artificial cultivated Radix gentianae was the highest, including 4.96% Gentiopicroside. The content of the underground part of 2-year-old Radix scutellariae harvested on 27 th August 2015 was the highest, including 14.99% Baicalin. The con-tent of Atractylodin in the Atractylodes chinensis was 0.23%, which was lower than the standard of Chinese Pharma-copoeia. Conclusion The method is simple, with accurate and reliable findings, which lays a foundation for the stan-dardized planting and harvest of the 5 traditional chinese medicinal materials.
