苯并芘诱导的小鼠腹主动脉瘤模型的建立 被引量:3

摘要 目的:建立苯并芘诱导的小鼠腹主动脉瘤模型,并与经典ApoE基因敲除(ApoE^(-/-))模型进行比较。方法:采用苯并芘高脂饲料喂养C57BL/6小鼠合并皮下注射血管紧张素II(Bap模型)诱导腹主动脉瘤的形成,造模第28天与ApoE^(-/-)模型比较腹主动脉瘤形成率、大体观的差别以及组织形态学的不同。结果:与正常对照组比较,苯并芘模型实验组小鼠腹主动脉表现为管腔扩张,直径增大(P<0.001)且未出现壁间血肿,成瘤率为40%;ApoE^(-/-)模型实验组小鼠腹主动脉直径明显增加(P<0.01)并伴有壁间血肿形成,成瘤率为60%。结论:与ApoE^(-/-)模型比较,苯并芘模型成瘤率虽然有所降低,但造模成本低廉同时模拟人类主动脉瘤的病理特征。 Objective:To establish mouse model of abdominal aortic aneurysm induced by benzopyrene and compare with the classi cal ApoE -/- model. Methods: Aortic aneurysm was induced by combination of 0.01 $ benzopyrene in high fat diet with angiotensin II treatment subcutaneously. The incidence,macroscopic and morphologic difference were compared with ApoE- /- model at the 28th day after modeling. Results:Compared with normal control group, the lumen diameter of benzopyrene group was increased signifi- cantly (P〈0. 001 ), there is no formation of intramural hematomal and the incidence of abdominal aortic aneurysm was 40 %;the lumen diameter of ApoE- /- group was also increased significantly (P〈0.0l)associated with formation of intramural hematoma,and the incidence of abdominal aortic aneurysm was 60%. Conclusion: Compared with ApoE- / -model, the incidence of benzopyrene group was lower,but it can simulate the pathological characteristics of human abdominal aortic aneurysm with a lower cost.
