不同浓度雌激素对急性期高血压脑出血患者的内皮祖细胞功能及衰老的影响 被引量:7

摘要 目的分析不同浓度的雌激素干预急性期高血压脑出血患者的内皮祖细胞(EPCs)后其功能变化及对衰老的影响。方法选取2015年3月至2016年6月在我院神经内科进行治疗的男性患者80例,平均分为5组,一组向其培养液中添加等量0.9%氯化钠注射液,作为对照组,3组分别添加浓度为10nmol/L、100nmol/L、1μmol/L的雌激素,作为雌激素组,最后一组先用磷脂酰肌醇-3-激酶(PI3K)抑制剂干预1h后,再用1μmol/L的雌激素干预。分别观察分析雌激素对EPCs的功能及衰老的影响。结果 EPCs形成集落的数量与雌激素的浓度呈正比(t=0.314,P=0.004);EPCs细胞的黏附能力、增殖能力、迁移能力随着雌激素浓度的增加而增加(P值分别为0.003、0.004、0.004);EPCs细胞的血管内皮生长因子(VEGF)蛋白质表达效果和抗衰老能力与雌激素浓度呈正比,差异均具有统计学意义(P均<0.05)。拮抗剂LY对雌激素有抑制作用。结论雌激素可以促进急性期高血压脑出血患者的EPCs集落,可增强其黏附能力、增殖能力、迁移能力,有利于EPCs VEGF蛋白质的表达,可以很好地抑制EPCs衰老。 Objective To investigate the effects of different concentrations of estrogen on endothelial progenitor cells(EPCs)in patients with acute hypertensive intracerebral hemorrhage(ICH).Methods Select 80 male patients who were treated in our hospital from March 2015 to June 2016.They were divided into five groups,agroup of medium saline solution added to it,as the control group,three groups were added at a concentration of 10nmol/L,100nmol/L,1μmol/L as estrogen,estrogen group,the last group with PI3 Kinhibitor 1h treated with 1μmol/L estrogen intervention.The effects of estrogen on EPCs function and aging were observed and analyzed.Results The concentration of EPCs was proportional to the number of colony formation and the estrogen(t=0.314,P=0.004);adhesion ability,proliferation ability and migration ability of EPCs cells increased with the concentration of estrogen;EPCs cells VEGF protein expression and the effect of antiaging ability and estrogen concentration differences were statistically significant(all P〈0.05).Antagonist LY inhibits estrogen.Conclusion Estrogen can promote the colony of EPCs in patients with acute hypertensive cerebral hemorrhage,can enhance the adhesion ability,proliferation ability and migration ability,is conducive to the expression EPCs VEGF protein,can effectively suppress the EPCs aging.
