冬凌草甲素下调NF-κB/COX-2降低人胃癌HGC-27细胞侵袭能力 被引量:9

Oridonin inhibits cell invasiveness through inhibiting the expression of NF-κB/COX-2 in HGC-27 gastric cancer cells

摘要 目的观察天然植物冬凌草提取物冬凌草甲素对胃癌HGC-27细胞侵袭能力的作用,并分析其可能的作用机制。方法体外培养HGC-27细胞,分别加入0、20、40、80μmol/L浓度的冬凌草甲素作用24 h,Transwell小室法检测细胞侵袭能力的改变;ELISA法检测细胞上清液中前列腺素E2(prostaglandin E2,PGE2)的含量;采用Western blotting检测环氧合酶-2(cyclooxygenase-2,COX-2)、p-NF-κB(p65)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达。结果经冬凌草甲素处理后,穿过Trasnwell小室基底膜的细胞数随着浓度的增加而逐渐减少,与对照组比较,差异有统计学意义(P<0.05);MMP-9表达下调,p-NF-κB(p65)、COX-2的表达及PGE2合成降低(P<0.05)。结论冬凌草甲素可能通过抑制p-NF-κB、COX-2、PGE2、MMP-9表达来降低HGC-27细胞侵袭能力。 Objective To explore the effect of oridonin on the cell invasiveness in HGC-27 gastric cancer cells,and weinvestigate the mechanisms. Methods The experiments were divided into two groups,including control group and drugtreated group. The control group received only the culture medium(oridonin 0 μmol/L). After treatment by oridonin in differentconcentrations(20,40 and 80 μmol/L) respectively for 24 h,the invasion of HGC-27 cells was tested by Transwell assay. Theexpression of PGE2 in the culture medium was detected by ELISA. Western-blotting were used for COX-2,p-NF-κB(p65)and MMP- 9 protein analysis. Results Compared with control group,the invasion of HGC- 27 cells was significantlydecreased in oridonin increased treatment group(P〈0.05). Oridonin inhibited the PGE2synthesis(P〈0.05). And the data ofwestern blot showed that oridonin decreased the expression of MMP- 9,COX- 2 and p- NF- κB(P〈0.05). Conclusion Oridonin inhibits the cell invasion in HGC- 27 cells,and the effects of antitumor may be associated with inhibition of theexpression of p-NF-κB,down-regulating the expression of COX-2 and PGE2 synthesis,thus down-regulating the expressionof MMP-9.
