4-硫胸苷与近紫外光协同抗癌辅助疗法研究进展

摘要 传统的癌症治疗方法包括手术、放射、化学药物治疗等,这些疗法虽经过数十年的发展和进步,效果仍不能令人完全满意。4-硫胸苷(S4TdR)作为一种核苷酸类似物,能够经嘧啶核苷酸补救合成途径替代普通胸苷靶向进入肿瘤细胞DNA链,并在近紫外光(UVA)照射下通过自身结构变化导致肿瘤细胞死亡。在多项体外研究中,S4TdR/UVA疗法被证实能够有效杀伤多种恶性肿瘤细胞,而对正常细胞无影响或杀伤能力弱。在动物模型中,S4TdR/UVA疗法的有效性和特异性得到了进一步的证实。对于多种生长于体表或管腔器官中的癌症,S4TdR/UVA有潜力作为一种精准的靶向疗法,辅助现有的传统抗癌方法,达到进一步提高综合治疗效果、改善患者预后的目的。因此,对S4TdR/UVA疗法的后续研究将具有重要意义。本文总结了近年来国内外S4TdR/UVA抗癌疗法的研究进展。 Traditional anti-cancer therapies includes surgery,radiotherapy and chemotherapy,etc.In spite of tens of years' development and progress,the effect of those therapies are still not satisfying.As a sulfur-containing nucleoside compound,4-thiothymidine(S4TdR)has similar structure and internal metabolism process compared with common thymidine(TdR),and a specific affinity to thymidine kinase(TK),which made it capable to be integrated into DNA molecules of Tumor cells.When irradiated under 335 nm Ultraviolet(UVA),S4 TdR can cause DNA chain breakage by its own conformational change,and further leads to specific cell death.Here we summarized the mechanism and research advances of S4TdR/UVA therapy.Since 2001,plenty research in both vitro and vivo confirmed that S4TdR/UVA as a safe,efficient,economic and targeting therapy for multiple types of cancer,especially in those grow on body surface or in lumen organs.To realize a better curative effect,and to improve the prognosis of cancer patients,S4TdR/UVA is hopeful to be applied to clinical practice to assist traditional anti-cancer therapies.Thus,we expect more profound and advanced studies to verify its feasibility.
