摘要
以35份马尾松种质资源为材料,研究马尾松嫩梢基因组DNA提取的最适方法;建立并优化马尾松ISSR反应体系;采用ISSR标记,结合聚类分析,对我国马尾松种质资源的遗传多样性进行了研究。结果表明:16条引物共扩增出153个条带,多态位点136个,每个引物检测到的条带数介于6~14之间;根据35份马尾松各资源的遗传系数进行聚类,可将35份资源划分为4个大类,即福建邵武、浙江仙居、江西德兴、福建连城各为1类,其余31份资源为1大类。
Using 35 accessions of Pinus massoniana as materials, optimum method of genomic DNA extracted from Pinus massoniana tender tip was studied, the ISSR reaction system was established and optimized,genetic diversity of Pinus massoniana was studied by ISSR markers and clustering analysis. The results showed that 16 primers amplified 153 bands among which had polymorphic loci 136,the percentage of polymorphic loci was 88.9%. The number of bands per primer detected among 6-14.According to the genetic coefficient, 35 resources could be divided into 4 categories, namely Fujian Shaowu ,Zhejiang Xianju, Jiangxi Dexing, Fujian Liancheng,the rest of the 31 resource for 1 categories.
