火麻仁木脂素酰胺类提取物对D-半乳糖胺盐酸盐致小鼠肝损伤的保护作用

Protective Effect of Lignanamides Extract from Fructus Cannabis on Hepatic Injury by D-galactosamine in Mice

目的观察火麻仁木脂素酰胺类提取物对D-半乳糖胺盐酸盐致小鼠急性肝损伤的保护作用。方法 KM种小鼠60只,按体质量随机分为正常对照组、D-半乳糖胺盐酸盐模型组、联苯双酯(150mg·kg^(-1))组,火麻仁木脂素酰胺类提取物高、中、低剂量组(180、120、60mg·kg^(-1))。连续给药7d后,腹腔注射10%D-半乳糖胺盐酸盐生理氯化钠溶液(10ml·kg^(-1))建立小鼠急性肝损伤模型,16h后,处死鼠取材,分别检测小鼠血清中谷丙转氨酶、谷草转氨酶活性,肝脏组织中脂质过氧化物丙二醛含量、超氧化物歧化酶活性,光学显微镜下观察肝组织病理学变化。结果 D-半乳糖胺盐酸盐模型组小鼠肝功能明显异常,血清中谷丙转氨酶、谷草转氨酶活性显著升高,肝脏组织变性、坏死严重。火麻仁木脂素酰胺类提取物各组血清中谷丙转氨酶、谷草转氨酶活性与模型组相比明显降低(P<0.01),肝组织匀浆检测发现超氧化物歧化酶活力增加,而脂质过氧化物丙二醛含量下降(P<0.01)。肝脏病理切片结果显示,火麻仁木脂素酰胺类提取物能明显改善受损组织的病理学变化。结论火麻仁木脂素酰胺类提取物对D-半乳糖胺盐酸盐致小鼠急性肝损伤具有较好的保护作用,其抗氧化能力可能是其保肝降酶的作用机制之一。

Objective To observe the protective effect of lignanamides extract from Fructus Cannabis （LFC） on acute hepatic injury induced by D-galactosamine（D-Galn） in mice. Methods According to body weight,sixty KM mice were pretreated with LFC at high dose （180 mg·kg^-1 ）, middle dose （120 mg·kg^-1） and low dose （60 mg·kg^-1） for 7 days. Mouse models of acute liver injury were established by the treatment with D-Galn. After 16 hours, the activities of aspartate aminotransferase and alanine amin- otransferase in serum,malonic dialdehgde levels and the activity of superoxide dismutase in the liver were measured by detection kits according to the manufacturers protocols.The histopathological changes of the liver were observed under a microscope.Results At each dose of LFC, the activities of alanine aminotransferase and aspartate aminotransferase in serum （ P〈0.01） and maleic dialdehyde level in the liver （ P 〈0.01） significantly reduced, but the activity of superoxide dismutase in the liver significantly increased com- pared with the D-Galn model group. Under the microscope, liver injury induced by D-Galn greatly decreased in each dose group of LFC.Conclusion LFC has protective effect on liver damage induced by D-Gain in mice in a dose-dependent manner. The ability of LFC to combat oxidation, may be one of the mechanisms.
