摘要
目的探讨建立改良的盲肠结扎穿刺法制备大鼠脓毒症模型。方法将20只雄性SD大鼠随机分为假手术组与脓毒症模型组,每组10只,分别进行假手术及盲肠结扎穿刺手术。各组大鼠分别于造模前、后不同时间点股静脉取血1ml,进行血常规和肌酐、尿素氮、谷丙转氨酶、谷草转氨酶检测,同时观察大鼠术后24h存活率。结果 24h生存率:假手术组大鼠为100%,模型组大鼠为20%;与假手术组相比,模型组大鼠白细胞计数术后2h显著性降低(P<0.05);肌酐、尿素氮水平6h显著升高(P<0.05);谷丙转氨酶与谷草转氨酶8h均显著升高(P<0.05,P<0.01)。结论改良的盲肠结扎穿刺法能建立稳定的大鼠脓毒症模型。
Objective To establish a rat sepsis model using advanced cecal ligation and puncture (CLP).Methods Twenty male SD rats were randomly divided into two groups: sham operation group and sepsis model group ( n = 10).A sepsis model was established according to the advanced CLP procedure, whereas sham group was sham operated.The blood samples of rats (1 ml each rat) were collected before and after the operation at different time points for determination the blood routine, creatinine, urea nitrogen,ALT and AST.Meanwhile, the survival rate 24 h after the operation was observed.Results The survival rate of sham group was 100% ,but 20% in sepsis model group.Compared to sham group, the white cell count of sepsis model group decreased significantly 2 h after the operation ( P 〈0.05);the levels of creatinine and urea nitrogen increased significantly from 6 h ( P 〈0.05),and the levels of ALT and AST increased significantly from 8 h ( P 〈0.05, P 〈0.01).Conclusion Advanced CLP can be used to establish a stable rat sepsis model.
