小檗碱吴茱萸碱配伍的降脂作用与上调LXRα和PPARγ蛋白表达的研究 被引量:4

摘要 目的:探讨小檗碱吴茱萸碱配伍对高脂血症大鼠肝脏LXRα和脂肪PPARγ蛋白表达的影响。方法:将60只大鼠随机分为空白组6只与模型组54只。建立高脂饮食诱导高脂血症大鼠模型,选取造模成功的48只大鼠随机分为8组(每组6只)后药物干预8周,检测各组大鼠的血清三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平以评价药物作用效果;以Western blot方法检测肝脏LXRα和脂肪PPARγ蛋白的表达。结果:与模型组相比,小檗碱吴茱萸碱配伍能够明显降低高脂血症模型大鼠血清TC、LDL-C水平(P<0.01),且降脂效果优于小檗碱或吴茱萸碱单用,能够升高血清HDL-C水平(P<0.05),但其作用弱于吴茱萸碱单用;小檗碱吴茱萸碱配伍能够明显上调模型大鼠肝脏LXRα和脂肪PPARγ的表达,且其作用强于小檗碱或吴茱萸碱单用(P<0.05)。结论:小檗碱吴茱萸碱配伍能够降低高脂饮食诱导的高脂血症大鼠血脂水平,其作用机制与其上调LXRα和PPARγ蛋白的表达有关。 Objective: To investigate the effect of a combination of berberine and evodiamine on the protein expression of liver X receptor alpha( LXRα) and peroxisome proliferator- activated receptor gamma( PPARγ) in rats with hyperlipidemia. Methods: A total of 60 rats were randomly divided into blank group( 6 rats) and model group( 54 rats). High- fat diet was given to establish a rat model of hyperlipidemia,and the 48 rats successfully established were randomly divided into 8 groups,with 6 rats in each group. After 8 weeks of drug intervention,the serum levels of triglyceride( TG),total cholesterol( TC),high- density lipoprotein cholesterol( HDL- C),and low- density lipoprotein cholesterol( LDL- C) were measured to evaluate the effect of drugs,and Western blot was used to measure the protein expression of LXRα and PPARγ. Results: Compared with the model group,a combination of berberine and evodiamine significantly reduced the serum levels of TC and LDL- C in rats with hyperlipidemia( P 〈0. 01) and had a better antihyperlipidemic effect than berberine or evodiamine alone; it also significantly increased the serum level of HDL- C( P 〈0. 05),but was inferior to evodiamine alone. A combination of berberine and evodiamine significantly upregulated the expression of LXRα and PPARγ in rats with hyperlipidemia and had a better effect than berberine or evodiamine alone( P 〈0. 05). Conclusion: A combination of berberine and evodiamine can reduce the blood lipid levels in rats with hyperlipidemia induced by high- fat diet,possibly by upregulating the protein expression of LXRα and PPARγ.
