结肠癌组织中microRNA-101 microRNA-132及COX-2的表达与病理相关性分析 被引量:3

摘要 目的分析结肠癌组织中miRNA-101、miRNA-132及COX-2的表达与病理相关性。方法收集2013年11月至2016年1月结肠癌病灶组织和癌旁正常组织各135例,应用反转录-聚合酶链式反应(RT-PCR)法检测组织中miRNA-101mRNA、miRNA-132mRNA和COX-2mRNA的表达,并分析各基因的相关性及与结肠癌临床病理特征的关系。结果与癌旁正常组织相比,结肠癌组织中的miRNA-101mRNA和miRNA-132mRNA的表达水平显著降低,COX-2mRNA显著上升,差异有统计学意义(P〈0.05);结肠癌组织中miRNA-101mRNA、miRNA-132mRNA和COX-2mRNA的表达与肿瘤的分化程度、TNM分期以及淋巴结转移有关;低中分化、TNM分期为Ⅲ期或Ⅳ期及淋巴结转移,结肠癌组织中miRNA-101mRNA和micrRNA-132mRNA的表达显著降低,COX-2mRNA的表达显著提高(P〈0.05);miRNA-101、miRNA-132与COX-2呈显著负相关(P〈0.05)。结论结肠癌组织中miRNA-101和miRNA-132表达下调,COX-2表达上调与结肠癌的发展进程有关,可作为结肠癌治疗的潜在靶点。 Objective To investigate the correlation of expression and pathology of miRNA-101, miRNA-132 and COX-2 in the patients with colon cancer. Methods The cancer and adjacent tissue of 135 patients with colon cancer in our hospital from November 2013 to January 2016 were involved in the study.RT-PCR was used to evaluate the expression of miRNA-101 mRNA, miRNA-132 mRNA and COX-2 mRNA in the tissue, and the correlation of each gene and the relationship between clinical pathological features of colon cancer was investigated. Results Compared with the adjacent tissues, the expression of miRNA-101 mRNA and miRNA-132 mRNA in the colon cancer tissues significantly decreased, the expression of COX-2 mRNA significantly increased, the difference was statistically significant ( P〈0.05 ) .The expression ofmiRNA-101 mRNA, miRNA-132 mRNA and COX-2 mRNA in the tissue was related to the degree of differentiation, TNM stage and lymph node metastasis.The expression of miRNA-101 mRNA and miRNA-132 mRNA significantly decreased in the tissues of medium or low degree of differentiation, TNM stage III IV or with lymph node metastasis, while COX-2 mRNA significantly increased (P〈0.05) ; which showed a significant negative correlation between miRNA-101, miRNA-132 and COX-2 ( P〈0.05 ) . Conclusion Down-regulated expression ofmiRNA-101 and miRNA-132 and up-regulated expression of COX-2 in the tissue is related to the process of colon cancer, and they can be used as potential targets of colon cancer therapy.
