小鼠糖尿病肾病足细胞NOD2 mRNA表达变化及其意义 被引量:5

摘要 目的观察糖尿病肾病(DKD)小鼠足细胞中细胞核苷酸结合寡聚化结构域蛋白2(NOD2)mRNA的表达变化,并探讨其意义。方法将分化完全的小鼠足细胞接种至6孔板,分为DKD组和NC组。DKD组分别加入25、50 mmol/L的膜过滤葡萄糖溶液刺激24 h制备DKD足细胞模型;NC组不给予糖刺激。采用实时荧光定量PCR法检测两组细胞中的NOD2 mRNA。选取上一实验中筛选出的最适浓度葡萄糖刺激足细胞制作DKD模型,分为胞壁酰二肽(MDP)组、shRNA组和模型组。MDP组给予2 g/m L的MDP刺激,shRNA组转染NOD2 shRNA,模型组常规培养,24 h后采用实时荧光定量PCR法检测各组细胞中的NOD2、nephrin、肿瘤坏死因子α(TNF-α)mRNA。结果 DKD组细胞中NOD2 mRNA相对表达量高于NC组,DKD组中50 mmol/L亚组NOD2 mRNA相对表达量高于25 mmol/L亚组(P均<0.05)。MDP组细胞中NOD2、TNF-αmRNA相对表达量高于模型组,nephrin mRNA相对表达量低于模型组(P均<0.05);shRNA组细胞中NOD2、TNF-αmRNA相对表达量低于模型组,nephrin mRNA相对表达量高于模型组(P均<0.05)。结论 DKD小鼠足细胞中NOD2 mRNA表达上调,NOD2 mRNA高表达时足细胞损伤加重,NOD2 mRNA低表达时足细胞损伤减轻;NOD2 mRNA表达变化可能与DKD的发病及病情进展有关。
