古法炮制地黄与市售地黄成分的比较

Component comparison of rehmanniae radix and rehmanniae radix praeparata between their ancient preparation and commercial medicines

通过高效液相色谱(HPLC)指纹图谱技术来研究古法炮制生、熟地黄药材与当前市售生、熟地黄的品质差异,为地黄药材质量标准的完善以及炮制方法的研究提供参考.样品采用甲醇提取,使用Welchrom C18色谱柱(4.6mm×250mm,5μm),乙腈-0.1%(体积分数)磷酸水溶液梯度洗脱,多波长紫外检测器检测,流速1.0mL/min,进样量20μL,柱温35℃.分别对生、熟地黄的HPLC指纹图谱的共有峰面积进行比对.古法炮制地黄与市售地黄所含成分有显著差异,古法竹刀切片制生地黄与古法酒蒸炮制熟地黄,其主要成分梓醇、毛蕊花糖苷含量普遍高于市售生、熟地黄;而5-羟甲基糠醛的含量远低于市售品.

The quality differences of rehmanniae radix and rehmanniae radix praeparata between their ancient preparation and commercial medicines were investigated by high performance liquid chromatography（HPLC）fingerprinting technology,providing reference for the improvement of their quality criterions and the study of their preparation processes of rehmanniae radix and rehmanniae radix praeparata.The samples were extracted with methanol and analyzed by HPLC.The chromatographic conditions were as follows：Welchrom C18（4.6 mm × 250 mm,5μm）column;acetonitrile-0.1% phosphate solution（V/V）was used as mobile phase for gradient elution;multi-wavelength UV detector;the flow rate was1.0mL/min and the column temperature was 35℃;the injection volume was 20μL.There were significant differences between the ancient preparation and commercial medicines through the comparison of their common peaks.The involved component contents,catalpol and acteoside of rehmanniae radix and rehmanniae radix praeparata from ancient preparation were higher than those of commercial medicines;while 5-HMF from ancient preparation were much lower than those of commercial medicines.The preparation processes of rehmanniae radix and rehmanniae radix praeparata are necessary to be further investigated and standardized to ensure their quality and efficacy.
