不同稀释液对人垂体泌乳素第3次国际标准品检测的影响

摘要 目的比较采用去离子水或含一定浓度蛋白的缓冲液作为稀释液,对人垂体泌乳素第3次国际标准品(84/500)测定的影响。方法采用磁微粒酶联免疫法、化学发光免疫法和时间分辨免疫荧光法等9家国产或进口试剂盒,测定不同稀释液配制的泌乳素国际标准品,计算实测值与理论值的比值。结果采用含一定浓度蛋白的缓冲液配制的国际标准品,实测值与理论值比值在0.93~1.20;采用去离子水或者不含蛋白的磷酸盐缓冲液配制,比值仅为0.25~0.46。结论泌乳素国际标准品的溶解必须选择含蛋白的缓冲液,且尽量采用各家试剂专用稀释液配制各浓度样本。 Objective To compare the effects of deionized water or buffer solutions with certain concentration of protein as sample dilution on the 3rd international standard of Prolactin,human,pituitary(84/500)measurement. Methods The PRL international standards dissolved by different solutions were examined by 9domestic or imported kits, including magnetic microparticle enzyme immunoassay, chemiluminescence immunoassay and time-resolved fluorescence immunoassay et al. Before the ratio of the measured value and the theoretical value was calculated. Results Using as PRL international standard buffer solution containing certain concentrations of protein,the ratio of the measured value and the theoretical value were 0. 93-1. 20.Using deionized water or the buffer without certain concentration of protein,the ratio were only 0. 25-0. 46.Conclusion Suitable dilution factors of protein must be selected for the dissolution of the international standard of prolactin. To generate reagent,the specific dilution buffer from manufacturerr should be used.
