白细胞介素-17A诱导脂肪细胞中炎症因子表达 被引量:2

INTERLEUKIN-17A DIFFERENTIALLY INDUCES INFLAMMATORY GENES EXPRESSION IN THE PREADIPOCYTES AND ADIPOCYTES

摘要 目的探讨IL-17A对脂肪细胞中炎症因子的诱导作用。方法诱导小鼠3T3-L1前脂肪细胞分化为脂肪细胞,20ng/mL IL-17A处理细胞2h,采用Real-time PCR检测处前脂肪细胞及脂肪细胞中炎症基因Tnf-α、Il-6、Il-1β、Cox-2、Il-17ra、Il-17rc、Ccl-2、Ccl-20、Cxcl1表达。结果脂肪细胞Tnf-α、Il-6、Cox-2、Ccl-20基因基础水平较前脂肪细胞增加(P<0.05),而Il-1β、Il-17ra、Il-17rc、Ccl-2基因则较前脂肪细胞减少(P<0.05),Cxcl1基因表达二者差异无统计学意义。给予外源性IL-17A处理,无论前脂肪细胞还是脂肪细胞,Tnf-α、Il-6、Il-1β、Cox-2、Il-17ra、Il-17rc、Cxcl1、Ccl-2、Ccl-20基因表达均较基础水平增加,且IL-17A对脂肪细胞中Tnf-α、Il-6、Cox-2基因的诱导作用强于前脂肪细胞。结论 IL-17A能够诱导前脂肪细胞和脂肪细胞中炎症基因差异表达,可能与肥胖及肥胖并发症发生有关。 Objective To investigate the induced effects of interleukin-17A(IL-17A)on inflammatory genes in preadipocytes and adipocytes.Methods Mouse 3T3-L1 pre-adipocytes were differentiated into adipocytes,followed with IL-17 A treatment and analysis for inflammatory genes expression.ResultsThe basal level of some genes such as Tnf-α,IL-6,Cox-2,Ccl-20 in adipocytes was higher than them in preadipocytes,while other genes such as IL-1β,IL-17 ra,Il-17 rc,Ccl-2 were lower compared to them in preadipocytes.All of the above inflammatory genes were increased after IL-17 A treatment.Conclusion IL-17 A differentially induced expression of inflammatory genes in pre-adipocytes and adipocytes,and may play an important role in the obesity and obesity-associated comorbidities.
