摘要
目的:建立反相高效液相紫外法同时测定刺五加叶中芦丁、金丝桃苷、槲皮素的含量测定方法。方法:采用Diamasil C_(18)色谱柱(200×4.6mm,5μm),以乙腈-0.3%磷酸水为流动相,流速为1m L·min^(-1),检测波长为360nm,柱温为30℃。结果:在40min内,刺五加叶中3个成分(芦丁、金丝桃苷、槲皮素)完全分离,峰面积的对数与进样量的对数呈现良好的线性关系(r=0.9995~0.9999);平均加样回收率在95.4%~99.7%,RSD为2.3%~3.3%。结论:该方法简便、迅速、可靠,适用于刺五加叶中黄酮类成分的含量测定。
Objective: To establish a reversed phase high performance liquid chromatography with ultro violet detector(RP-HPLC-UV)method for the simultaneous determination of three compounds in A. senticocus leaves. Methods: The Diamonsil C(18)column(250 mm × 4.6 mm, 5μm)was used for separated and analysis using gradient elution with a mobile phase of acetonitrile as solvent A and 0.3%(v/v)phosphoric acid in water as solvent B, the flow rate was1.0 m L·min^-1, detection wavelength was 360 nm and the column temperature was maintained at 30 ℃. Results: The three compounds(rutin, hyperin, quercetin)were completely separated within 40 min. The regression equation showed good liner relationship between logarithm of the peak area and logarithm of the content of each compound(r = 0.9995-0.9999). the average recoveries was 96.7%-99.1% and RSD was 2.3%-3.3%. Conclusion: The method is simple, fast and reliable, which can be used to determine the contents of three compounds in A. senticocus leaves.
