胡桃楸总鞣质的小鼠体内抗肿瘤作用研究 被引量:3

摘要 目的研究胡桃楸总鞣质的体内抗肿瘤活性。方法采用70%丙酮超声提取胡桃楸中总鞣质。采用荷H22肝癌实体瘤小鼠模型评价胡桃楸的抗肿瘤活性。将50只ICR小鼠随机分为正常组,模型组,环磷酰胺阳性对照组,总鞣质低、高剂量组,每组各10只。正常组小鼠右腋下接种PBS(0.2 m L/只),其余各组小鼠均于右腋下接种等体积的小鼠H22肝癌细胞悬液(2×10~6个/m L)。接种24 h后给药,总鞣质低(0.09 g/kg)、高剂量(0.18 g/kg)组分别灌胃给予低浓度和高浓度的总鞣质提取物,阳性对照组腹腔注射环磷酰胺生理盐水溶液(0.03 g/kg),正常组和模型组灌胃给予等体积的蒸馏水。连续给药10 d后,处死小鼠,以瘤重和抑瘤率为指标研究胡桃楸总鞣质对H22肝癌实体瘤的抑制作用,以免疫器官指数、外周血象和肝功能为指标研究胡桃楸总鞣质对荷瘤机体生存质量的影响。结果胡桃楸总鞣质的得率为5.07%,纯度为92.8%。与模型组比较,胡桃楸总鞣质低、高剂量组肿瘤重量明显降低(P<0.05),均显示较明显的抗肿瘤作用,抑瘤率分别为34.22%和36.92%。在抑制肿瘤生长的同时,胡桃楸总鞣质未显示环磷酰胺所致的体重降低、免疫抑制和杀伤白细胞等副作用。结论胡桃楸总鞣质具有较好的抗肿瘤作用,且无免疫抑制和杀伤白细胞等副作用。 Objective To evaluate the in vivo anti-tumor activity of the total tannins from Juglans mandshurica. Methods Total tannins from Juglans mandshurica was extracted by 70% acetone. Mice bearing murine-hepatocarcinoma (H22) model was established to evaluate the anti-tumor activity. Totally 50 ICR mice were randomly divided into five groups with 10 mice in each group: normal control group, model control group, cyclophosphamide group (as positive control), and total tannins low, high dose group. The normal control group was subcutaneously injected with PBS (0.2 mL per mouse), while the other four groups were subcutaneously injected with murine-hepatocarcinoma (H22) cell suspen- sion (2xl0a/mL). 24 h after injection, total tannins low, high dose group was intragastrieally administered with total tan- nins at low (0.09 g/kg) and high dosage (0.18 g/kg). Cyelophosphamide group was given with cyclophosphamide (0.03 g/kg) by intraperitoneal injection. Normal and model control groups were intragastrically administered with water. After continuous administration for 10 d, killed mice, tumor weight and tumor inhibition ratio were obtained for the evaluation of the anti-tumor activity. Immune organ index, hematological parameters and Hepatic functions were determined to estimate the protective effects of total tannins on normal functions of H22 tumor bearing mouse. Results The yield of tannins from Juglans mandshurica was 5.07%, and the purity of the tannins was 92.8%. Total tannins from Juglans mandshurica showed inhibitory effect on the growth of tumor tissues. The inhibitory ratio of total tannins low and high dose group were 34.22% and 36.92%, respectively. Total tannins from Juglans mandshurica did not exhibit side effects,which are typical of cyclophosphamide, such as loss of body weight, immune suppression and killing white blood cell. Conclusion Total tannins from Juglans mandshurica was effective in anti-tumor, and did not ex- hibit side effects of immune suppression and killing white blood cell.
