急性宫颈扩张对脊髓TRPA1表达的影响

摘要 目的探讨急性宫颈扩张(UCD)后胸腰段脊髓瞬时受体电位通道蛋白(TRPA1)表达的变化。方法成年雌性大鼠60只,随机分为5组:假手术组和UCD25、UCD50、UCD75、UCD100组。全凭吸入麻醉下,除S组外,各组大鼠分别采用25、50、75、100 g的砝码进行持续10 s的宫颈扩张,重复1次,间隔5 min;假手术组不作扩张。记录UCD时大鼠腹直肌肌电图及心率。待UCD结束后10 min处死大鼠,分别运用实时荧光定量PCR及Western blot法测定脊髓胸腰段TRPA1的变化。结果与假手术组相比,接受不同张力UCD的各组大鼠腹直肌肌电活动及胸腰段脊髓TRPA1表达均明显增加(P<0.05)。组间两两比较:与UCD25组或UCD50组相比,UCD75组和UCD100组肌电图活动及脊髓TRPA1表达明显增加,差异有统计学意义(P<0.05);但UCD25组与UCD50组间以及UCD75组与UCD100组之间差异无统计学意义(P>0.05);而且急性UCD后胸腰段脊髓TRPA1的表达变化与UCD对腹直肌肌电图的影响趋于一致。结论胸腰段脊髓TRPA1可能参与了急性UCD相关内脏痛的形成。
