摘要
背景 NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor pyrin domain containing 3, NLRP3)炎性体由NLRP3、凋亡相关斑点样蛋白(apoptosis-associated speck-like protein containing a CARD, ASC)和酶解活化半胱氨酸蛋白水解酶-1(cysteinyl aspartate specific proteinase-1, caspase-1)相互结合形成,通过活化caspase-1来间接调控IL-1β的成熟和分泌,在细胞质内发挥对外源性微生物或内源性危险信号感受器的作用。 目的 对NLRP3炎性体的调控机制研究进行综述。 内容 分别描述热蛋白结构域(pyrin domains, PYD)、泛素化、一氧化氮(nitric oxide, NO)、干扰素(interferon, IFN)、线粒体、活性氧(reactive oxygen species, ROS)等对NLRP3的调节机制。 趋向 为NLRP3相关疾病的诊疗提供参考。
Background NOD-like receptor pyrin domain containing 3(NLRP3) inflammasomes are multimeric protein that consists of NLRP3, apoptosis-associated speck-like protein containing a CARD(ASC) and cysteinyl aspartate specific proteinase-1(caspase-1). NLRP3 is an important regulator of caspase-1, the enzyme that processes the immature form of IL-1β into the active protein. IL-1β act as a receptor of exogenous microorganisms and endogenous danger signas. Objective The purpose of this review is to focus on how NLRP3 inflammasomes regulate inflammatory disease through negative regulation. Content To explain the roles of pyrin domains, ubiquitination, nitric oxide, interferon, reactive oxygen species in regulating effect of NLRP3 inflammasomes. Trend To provide some new ideas of the diagnosis and therapy for NLRP3 inflammasomes-related diseases.
