葡萄籽原花青素缓解硝基苯酚诱导的免疫毒性 被引量:2

摘要 3-甲基-4-硝基苯酚(PNMC)是重要的汽车尾气有效活性成分,对小鼠脾淋巴细胞具有免疫毒性作用。作为食物多酚类物质,原花青素具有抑制各种毒性作用,而葡萄籽富含原花青素。用细胞活力分析和ELISA实验,研究葡萄籽原花青素对PNMC免疫毒性影响结果表明:葡萄籽原花青素显著抑制PNMC对脾淋巴细胞增殖的毒性作用,缓解PNMC对脾T淋巴细胞分泌白细胞介素2、4及颗粒酶B功能的损伤作用。测定OH·、SOD、GSH-Px和MDA指标的抗氧化试验结果表明:葡萄籽原花青素缓解PNMC对脾淋巴细胞的免疫毒性作用与其抗氧化作用具有密切关系。 3-methyl-4-nitrophenol( PNMC),as a kind of important component of vehicle emissions,has been shown to confer toxicity in splenocytes. Proanthocyanidin,as a dietary polyphenols,has been shown to confer inhibitory activity against a variety of toxins,and grape seed is rich in proanthocyanidins. The effects of grape seed proanthocyanidin on immunotoxicity induced by PNMC in murine splenic lymphocytes were investigated by Cell Viability Assay and ELISA,and the results showed that grape seed proanthocyanidin significantly enhanced proliferation of splenocytes exposed to PNMC,as well as the production of T cell-related cytokines and granzymes including interleukin-2,interleukin-4,and granzyme-B in the cells exposed to PNMC. These effects were associated with a decrease in oxidative stress,as evidenced by changes in OH·,SOD,GSH-Px,and MDA levels.
