摘要
目的:探讨分子生物化学实验室内免疫组织化学染色中抗原修复的最佳方式。方法:采用高温高压修复法和微波修复法对病理科采用的四项免疫组织化学标记(ER、PR、C-erb B-2及E-Cadherin)进行染色对比以获取最佳抗原修复方法,在显微镜下评判其统计阳性率及阳性程度对两种方法进行比较。结果:ER、PR、C-erb B-2及ECadherin四项标记在高温高压抗原修复法与微波抗原修复法均具有较高的阳性率,在阳性程度、显色强度及背景清晰度方面高温高压抗原修复法要优于微波抗原修复法。结论:高温高压抗原修复法与微波抗原修复法均是简便实用的方法,两者相比高温高压抗原修复法具有更强适用性。
Objective:To discuss molecular biochemistry laboratory undergraduate course room immunohistochemical staining in antigen repair the best way. Methods:The four immunohistochemical markers( ER,PR,C- erb B- 2 and E- cadherin)were stained and contrasted in the pathology by high temperature and high pressure repair method and microwave repair method to obtain the best antigen retrieval method. Its statistical positive rate and positive degree of two methods were compared and evaluated under the microscope. Results:ER,PR,C- erb B- 2 C and E- Cadhrin four tags in high temperature and high pressure antigen repairing method and the microwave antigen repair method has higher positive rate,background of positive degree,color depth and clarity in high temperature and high pressure antigen repairing method is superior to the microwave antigen repair method. Conclusion:High temperature and high pressure antigen repairing method and the microwave antigen repair method is simple and practical method,both,compared to high temperature and high pressure antigen repair method has better applicability.
