摘要
[目的]分离鉴定寒地水稻纹枯病病原菌,为寒地水稻纹枯病抗性育种提供技术支持和理论依据。[方法]采集水稻纹枯病病株并分离培养病菌,采用柯赫氏检验法和ITS鉴定的方法,对分离菌株进行形态观察和分子生物学鉴定。[结果]将分离获得病菌以牙签嵌入法接种水稻植株,植株呈现水渍状典型纹枯病病斑。利用ITS1和ITS4引物扩增病菌DNA,测序结果拼接后与NCBI上已知序列比对,结果表明该菌与登录号为No.KT_362083.1的Rhizoctonia solani(立枯丝核菌)有97%的同源性。[结论]形态学特征鉴定与分子生物学鉴定结果一致,该菌为立枯丝核菌,为寒地水稻纹枯病致病菌。
[ Objective ] Strain of rice sheath blight in cold region was isolated, and identificated in order to provide technical support and theoret- ical basis for rice sheath blight resistance breeding. [ Method] Diseased plants from rice sheath blight were collected,and strain was isolated and cultured. Morphological observation and molecular identification on strain were carried out by using the method of Koch's test and ITS identifica- tion. [ Result] The isolated strain "inoculated rice plants with toothpick embedding method, the plants showed hygrophanous typical sheath blight spots. ITS1 and I'lS4 primers were used to amplify the strain DNA, sequencing results were compared with known sequences on NCBI, Blast searches results showed a high identity with reference sequences (ITS:97%). Representative sequences of both regions were deposited to GenBank (ITS:Accession No. KT362083.1 ). [ Conclusion ] Morphological and molecular results confirm this strain as Rhizoctonia solani, the strain is the pathogens of rice sheath blight.
