摘要
目的:研究大孔树脂纯化杨梅叶原花色素的条件。方法:通过静态实验,确定X-5、AB-8、NKA-9、H-103、HP-20等5种大孔树脂对杨梅叶原花色素的影响,以确定最佳的大孔树脂种类,以及研究时间、p H值、温度对杨梅叶原花色素吸附率的影响和洗脱剂、时间、温度对杨梅叶原花色素解吸率的影响。结果:供试的5种大孔树脂中,H-103树脂吸附和洗脱效果较好。在杨梅叶原叶液浓度(以吸光度计)A=1.601、振荡100 min、p H=1,45℃时,吸附率较佳。最佳解吸条件为以100%丙酮为洗脱剂,在40℃下振荡20 min。结论:试验结果表明优选出的纯化工艺提取率较高,值得推广。
Objective:To study the purification condition of proanthocyanidins from leaves of myrica rubra sieb.Methods:By static experimental methods,to make sure the influence of X-5,AB-8,NKA-9,H-103 and HP-20 macroporous resin on proanthocyanidins from leaves of myrica rubra sieb and optimun macroporous resin as well as study the effect of time,pH,and temperature on the adsorption of proanthocyanidins from leaves of myrica rubra sieb and study the effect of eluent,time and temperature on the desorption of proanthocyanidins from leaves of myrica rubra sieb.Results:Among the 5 macroporous resin tested,H-103 resin was better in adsorption efect and desorption effect than the others.As the following conditions:at concentration of leaves original solution(to represent with absorbance) was 1.601,oscillated 100 min,p H = 1,temperature 45℃,the adsorption effect is better.The optimum conditions of desorption effect are:at 40℃,oscillated 20 min by using 100% acetone desorption.Conclusion:The experimental results show that the optimized extraction purification is more reliable and worth popularizing.
