摘要 目的 研究细胞间粘附分子-1)ICAM-1)在蛛网膜下腔出血(SAH)后脑血管壁内的表达变化规律及其与脑血管痉挛(CVS)的关系。方法 应用免疫组织化学及逆转录-聚合酶链式反应(RT-PCR)等方法,检测大鼠2次注血SAH模型基底动脉壁内ICAM-1在蛋白质和mRNA水平上的表达。结果 在大鼠脑池内注血后第3d,基底动脉血管腔狭窄已非常明显,第5d达到高峰。注血后第1d,ICAM-1表达开始上调,主要在内膜及内膜下,第3d表达明显增强并持续到第5d。ICAM-1在蛋白和mRNA水平上表达基本一致,且在时程上的变化与CVS的程度密切相关。结论 由ICAM-1介导的血管壁炎症反应可能在CVS发生和发展过程中起重要作用。 Objective To study the expression of intercellular adhesion molecule-1 (ICAM-1) on the cerebral arteries following subarachnoid hemorrhage (SAH) in rats and its relationship with cerebral vasospasm (CVS). Methods Immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) were used to examine the expression of ICAM-1 protein and ICAM-1 mRNA in the basilar artery (BA) walls of rat SAH model. Results The BA vasospasm was obvious on day 3 after blood was injected into rat cistern, and peaked on day 5. There was expression of ICAM-1 in the BA walls accompanying with vasospasm. Up-regulation of ICAM-1 expression started on day 1, mainly in intima and subintima, became more stronger on day 3 and remained up on day 5. The level of protein and mRNA of ICAM-1 were mostly identical, and they significantly correlated with the degrees of CVS. Conclusion The results suggest that the inflammatory reaction in vessel walls mediated by ICAM-1 may play an important role in the development of CVS after SAH.
