摘要
目的探讨WASP家族Verprolin同源蛋白1(WAVE1)和高迁移率族蛋白-1(HMGB1)在兔尿源性脓毒血症肺组织中表达上调中的关系。方法 30只健康雄性家兔随机分为假手术组(Sham组)和尿源性脓毒血症组(US组)。采用肾盂内高压法制备家兔尿源性脓毒血症模型。免疫组织化学染色SABC法分别于术后12 h、24 h、36 h检测家兔肺组织WAVE1蛋白表达水平及肺组织中HMGB1含量及HMGB1 mRNA表达。结果与Sham组比较,US组肺组织HMGBl含量、HMGBl mRNA表达活性升高(P<0.05);免疫组织化学结果显示,US组肺组织中WAVE1 80.00%(12/15))呈强阳性表达,在Sham组肺组织中,WAVE1 86.67%(13/15))呈低表达或不表达,差异具有统计学意义(P<0.05)。结论尿源性脓毒血症急性肺损伤中WAVE1与HMGB1可能具有相关性。
Objective To explore the relationship between WASP-family verprolin-homologous protein 1 and high mobility group box 1 in lung tissues in septic rabbits.Methods Thirty healthy male rabbits were randomly divided into 2 groups: sham operation group (Sham group), urosepsis group (US group). The model of urpsepsis was induced by acute pelvic obstruction in high pressure. SABC immunohistochemistry was done respectively to determinate WAVE1 protein expression levels, HMGB1 content and HMGB1 mRNA expression in lung tissues at 12 h, 24 h and 36 h after operation. Compared with Sham group, HMGB1 content and HMGB1 mRNA expression were significantly increased in US group (P〈0.05). WAVE1 was strongly positively expressed in 80% of US group, while it was weakly positive or negative expressed in 86.67% of Sham group. The differences between the two groups were statistically significant (P〈0.05). Conclusions WAVE1 potentially involved in the up-regulation of HMGB1 expression in lung tissues in septic rabbits.
