摘要
目的研究香菊胶囊溶液对单纯疱疹病毒2型的抗病毒作用,为生殖器疱疹的治疗寻找新的治疗手段。方法以Vero细胞为宿主细胞,阿昔洛韦为阳性对照药物,通过观察细胞病变效应(CPE)、噻唑蓝(MTT)比色法检测细胞存活率和病毒抑制率来评价香菊胶囊溶液体外对单纯疱疹病毒2型(HSV-2)的抑制作用。结果香菊胶囊溶液对Vero细胞的半数毒性浓度(TC_(50))为12.5mg/m L,药物和HSV-2共同作用2h后、病毒感染2h后加入药物、预防给药途径三种条件下对HSV-2的半数有效浓度(IC_(50))分别为1.901mg/m L,1.871mg/m L和1.942mg/m L,治疗指数(TI)分别为6.58,6.68和6.44,其抗病毒效果与阿昔洛韦差异无统计学意义(P>0.05)。结论香菊胶囊溶液在体外对HSV-2直接灭活作用明显,且对HSV-2的吸附和穿入细胞均有抑制作用,具有很好的体外抗疱疹病毒作用。
Objective To evaluate the antiviraI activity of Xiangju capsule solution against herpes simple virus 2（HSV-2）in vitro.Methods The Vero cell was used to detect the antiviral effect of Xiangju capsule solution compared with acyclovir in vitro.The studied were conducted by assaying cytopathogenic effect（CPE）for protection and MTT test for viral inhibitory rate.Results The TC50 value of Xiangju capsule solution was 12.5mg/mL. Of these three conditions,combination Xiangju capsule solution and HSV-2 lasted for 2 hours,2 hours after infection then given the capsule,and use of preventive medicine,IC50 value was analysed. The variation of the half effective concentration against HSV-2 was 1.901mg/mL,1.871mg/mL and 1.942mg/mL,respectively. And the therapeutic index（TI）value was 6.58,6.68 and 6.44 respectively.Conclusion The results indicate that Xiangju capsule solution displays inhibitive effect on HSV-2 directly and has preferable influence on virus attachment and virus penetration.
