血清miRNA-146、miRNA-195检测在糖尿病视网膜病变中的临床应用 被引量:9

摘要 目的探讨血清微小RNA(miRNA)-146、miRNA-195检测在糖尿病视网膜病变(DR)中的临床应用价值。方法收集91例糖尿病视网膜病变患者的血清及临床资料,并从正常人群中随机抽取45例志愿者。采用实时荧光定量PCR(RT-PCR)技术检测血清miRNA-146、miRNA-195的表达。将糖尿病视网膜病变患者分为非增殖性糖尿病视网膜病变组(NPDR)和增殖性糖尿病视网膜病变组(PDR),与对照组比较,观察血清miRNA-146、miRNA-195表达水平的变化与DR之间的关系。结果病例组血清miRNA-146、miRNA-195表达与对照组比较分别上调和下调,差异均有统计学意义(P<0.01);miRNA-146在NPDR组的表达水平低于PDR组(P<0.01),miRNA-195的表达水平则高于PDR组(P<0.01)。NPDR组和PDR组miRNA-146、miRNA-195表达均呈负相关(r=-0.687,-0.692,P<0.01)。NPDR组和PDR组中miRNA-146、miRNA-195的表达水平分别和糖化血红蛋白(Hb Alc)、尿白蛋白/肌酐及空腹血糖(FPG)水平具有相关性(r=0.532,0.610,0.587,P<0.01);(r=-0.538,-0.627,-0.567,P<0.01)。结论检测血清miRNA-146、-195表达可预测DR的发生及判断其危险程度,估计病情的发展状况。 Objective To explore the clinical application value of the levels of serum miRNA-146,miRNA-195 in diabetic retinopathy. Methods The plasma and clinical data from 91 cases of the patients with DR were collected,38 cases of volunteer blood plasma from a random sample of the general population were extracted. The Expression levels of miRNA-146,miRNA-195 were quantitatively detected by RT-PCR method. The patients with diabetic retinopathy were divided into the non-proliferating diabetic retinopathy group( NPDR) and proliferating diabetic retinopathy group( PDR). The relationship between the changes of the expression of the levels of the serum miRNA-146,miRNA-195 and DR were observed. Results In the case group,the levels of serum miRNA-146,miRNA-195 raised and lowered respectively,compared with that in control group with significant difference( P〈0.01). The expression levels of miRNA-146 in NPDR group were lower than that in PDR group( P〈0.01). The expression of the miRNA-195 were higher than that in PDR group( P〈0.01). The levels of miRNA-146,miRNA-195 were negatively correlated in both NPDR and PDR group( r=0. 687,-0. 692,P〈0.01). The expression level of miRNA-146 and miRNA-195 respectively had correlation with the levels of glycosylated hemoglobin( HbA1c),urine white silkworm / creatinine and fasting plasma glucose( FPG) in NPDR and PDR group( r=0. 532,0. 610,0. 587,P〈0.01; r =-0. 538,-0. 62,-0. 56,P〈0.01). Conclution Detection of the expression of serum miRNA-146,miRNA-195 can predict the occurrence of DR and help to determine its dangerous degree and estimate the development of the disease.
