香芹酚对大鼠痛风性关节炎中血清炎性因子及软骨细胞凋亡的影响 被引量:13

The effects of carvacrol on inflammatory cytokines and the apoptosis of cartilage in rats with gouty arthritis

摘要 目的探索香芹酚(CAR)对痛风性关节炎(GA)大鼠炎性因子的影响,以及对膝关节软骨细胞凋亡的影响。方法尿酸钠诱导大鼠痛风性关节炎模型,60只大鼠随机分为5组,A组(正常组),B组(模型组),C组(香芹酚10mg/kg),D组(香芹酚25 mg/kg)和E组(香芹酚50 mg/kg),每组12只。A、B组给予等量生理盐水处理,连续给药14 d,C、D、E分别在GA造模后给予香芹酚10 mg/kg、25 mg/kg、50 mg/kg,每日一次,灌胃喂食。7 d和14 d分别观察各组大鼠踝关节肿胀程度,并利用ELISA法检测各组大鼠血清中肿瘤坏死因子α(TNF-α),白细胞介素1β(IL-1β),前列腺素E2(PGE2),转换生长因子β1(TGF-β1)。14 d取踝关节软骨,利用Western blot检测软骨Bax和Bcl-2蛋白表达。结果 7 d和14 d B组大鼠踝关节肿胀程度显著高于A组,而C、D、E组明显低于B组(P均<0.05),且呈剂量依耐性。在给药第7 d和14 d,B组大鼠血清TNF-α,IL-1β,PGE2水平显著高于A组(P均<0.05),而C、D、E组血清TNF-α,IL-1β,PGE2水平明显低于B组(P均<0.05),且呈剂量依耐性。而7 d和14 d B组大鼠血清TGF-β1水平显著低于A组(P均<0.05),而C、D、E组血清TGF-β1水平明显高于B组(P均<0.05),且呈剂量依耐性。14 d B组大鼠踝关节软骨Bax蛋白表达明显高于A组,而C、D、E组明显低于B组(P均<0.05),B组Bcl-2蛋白表达明显低于A组,而C、D、E组明显高于B组(P均<0.05),且呈剂量依耐性。结论香芹酚能显著抑制痛风性关节炎大鼠踝关节肿胀度,抑制炎症反应以及减轻软骨细胞的凋亡,且香芹酚的作用呈剂量依耐性。 Objective To explore the effects of carvacrol on inflammatory cytokines in serum of gouty arthritis rats,and to study its effects on the apoptosis of cartilage. Methods Gouty arthritis model were induced with sodium urate,and 60 SD rats were randomly divided into 5groups,group A( The control group),group B( The model group),group C( carvacrol 10 mg / kg),group D( carvacrol 25 mg / kg),group E( carvacrol 50 mg/kg). Ankle swelling was observed at 7 d and 14 d after medical treatment. Meanwhile,the inflammatory cytokines in serum such as tumor necrosis factor α( TNF-α),interleukin 1β( IL-1β),prostaglandins E2( PGE2) and transforming growth factor β1( TGF-β1) were also detected by elisa at 7 d and 14 d after medical treatment. Besides,the protein expression of Bax and Bcl-2 were detected by western blot at 14 d after medical treatment. Results The levels of ankle swelling in group B were significantly higher than those in group A at 7 d and14 d after medical treatment( P〈0.05),while,those in group C,D,E were effectively lower than that in group B( P〈0.05). Meanwhile,the serum level of TNF-α,IL-1β,PGE2 in group B were significantly higher than those in group A at 7 d and 14 d after medical treatment( P〈0.05),while,those in group C,D,E were effectively lower than that in group B( P〈0.05),meanwhile,TGF-β1 activity in group B was markedly lower than that in group A,while,those in group C,D,E were effectively higher than that in group B( P〈0.05). Besides,the protein expression of Bax in group B was markedly higher than that in group A,while,those in group C,D,E were effectively lower than that in group B( P〈0.05),and the expression of Bcl-2 in group B were markedly lower than that in group A,while,those in group C,D,E were effectively higher than that in group B( P〈0.05). Conclusion Carvacrol could effectively mitigate the ankle swelling level of gouty arthritis rats,and it could also suppress the serum inflammation and the apoptosis of cartilage,and the effect of carvacrol is dose-dependent.
