脂质体阿霉素用于治疗小鼠淋巴瘤的效果研究 被引量:3

摘要 目的探讨脂质体阿霉素用于治疗小鼠淋巴瘤的实验效果。方法选取C57BL/6淋巴瘤模型小鼠120只,采用随机数字表法分为空白组(等量生理盐水)、游离阿霉素组(1 mg/kg游离阿霉素,连续3次给药)、空白脂质体+游离阿霉素组(1 mg/kg游离阿霉素+1.2 mmol/kg空白脂质体,连续3次给药)、脂质体阿霉素组(1 mg/kg脂质体阿霉素,连续3次给药)。观察各组小鼠的60 d生存率、中位生存时间,各组小鼠接种肿瘤细胞后第3、6、12、24天的肿瘤体积。采用CD31染色检测各组小鼠的肿瘤组织微血管密度(MVD)、TUNEL染色检测肿瘤细胞凋亡率。结果游离阿霉素组、空白脂质体+游离阿霉素组、脂质体阿霉素组的肿瘤体积在第6、12、24天均小于空白组(P<0.05);脂质体阿霉素组的肿瘤体积在第3、6、12、24天均小于空白组、游离阿霉素组、空白脂质体+游离阿霉素组(P<0.05);脂质体阿霉素组60 d生存率为40%,明显高于游离阿霉素组、空白脂质体+游离阿霉素组、空白组的生存率0(P<0.05);脂质体阿霉素组的中位生存时间大于游离阿霉素组、空白脂质体+游离阿霉素组(P<0.05);脂质体阿霉素组的MVD值小于游离阿霉素组、空白脂质体+游离阿霉素组(P<0.05),肿瘤细胞凋亡率大于游离阿霉素组、空白脂质体+游离阿霉素组(P<0.05)。结论脂质体阿霉素用于治疗小鼠淋巴瘤能够更加有效地抑制肿瘤生长、促进细胞凋亡。 Objective To investigate the effects of liposome adriamycin on mice with lymphoma. Methods 120 C57BL/6 lymphoma model mice were selected,randomly divided into control group( saline),free doxorubicin group( 1 mg / kg free doxorubicin,3 consecutive administration),blank liposomes and free adriamycin group( 1 mg / kg free doxorubicin + 1. 2 mmol / kg blank liposomes,3 consecutive administration),liposomal doxorubicin group( 1mg / kg liposomal doxorubicin,3 consecutive administration). The 60 d survival rate,median survival time and tumor volume of 3,6,12,24 d after inoculation of tumor cells in each group were observed; CD31 staining was used to detect the tumor tissue microvessel density( MVD) and TUNEL staining to detect the apoptosis rate of tumor cells. Results The tumor volumes of free doxorubicin group,blank liposome + free doxorubicin group and doxorubicin group in 6,12 and 24 d were all smaller than that in the blank group( P〈0.05). The tumor size of doxorubicin group in 3,6,12,24 d were smaller than those in the blank group,free doxorubicin group,blank liposome + free doxorubicin group( P〈0.05). The 60 d survival rates of free doxorubicin group,blank liposome + free doxorubicin group and blank group were0,lower than doxorubicin group( 40%)( P〈0.05). The median survival time of doxorubicin group was greater than that of free doxorubicin group,blank liposome + free doxorubicin group( P〈0.05). The MVD value of doxorubicin group was lower than those of free doxorubicin group,blank liposome and free doxorubicin group( P〈0.05),the apoptosis rate of tumor cells was higher than those of free doxorubicin group,blank liposome and free doxorubicin group( P〈0.05). Conclusion Liposome doxorubicin can be more effective in the treatment of lymphoma in mice to inhibit tumor growth and promote cell apoptosis.
