摘要
目的:探讨长链非编码RNA SPRY4-IT1在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织和细胞系中的表达水平,以及SPRY4-IT1对NSCLC细胞增殖和侵袭功能的影响及分子机制。方法:用实时定量PCR技术检测SPRY4-IT1在NSCLC组织及细胞系中的表达水平。将p CDNA-SPRY4-IT1转染SPC-A1和A549细胞,过表达SPRY4-IT1,用MTT法和克隆形成实验检测SPRY4-IT1对SPC-A1和A549细胞增殖和克隆形成能力的影响。Transwell实验评价SPRY4-IT1对细胞迁移、侵袭能力的影响。q PCR和Western blot检测过表达SPRY4-IT1对上皮-间质转化(epithelial-mesenchymal transition,EMT)标志物上皮性钙粘蛋白(E-cadherin)、波形蛋白(Vimentin)表达水平的影响。结果:SPRY4-IT1的表达在NSCLC组织和细胞系中出现显著下调(P<0.01)。过表达SPRY4-IT1能降低SPC-A1细胞和A549细胞的增殖和侵袭能力。过表达SPRY4-IT1显著上调E-cadherin的水平、抑制Vimentin的表达,从而影响肿瘤细胞EMT过程。结论:长链非编码RNA SPRY4-IT1表达下调可能通过调控EMT机制促进NSCLC细胞的增殖和侵袭。
Objective:To study the long non- coding RNA SPRY4- IT1 in non- small cell lung cancer( NSCLC) expression in tissues and cell lines,and the mechanisms how SPRY4- IT1 effects on cell proliferation and invasion. Methods:Quantitative PCR was used to detect the expression of SPRY4- IT1 in NSCLC tissue and cell lines. Transfect p CDNA- SPRY4- IT1 to SPC- A1 and A549 cells to overexpress SPRY4- IT1 and then evaluate the proliferation and clonogenic activity using clonogenic assay and MTT method. Transwell experiment was used to assess the effects of SPRY4- IT1 on the invasion of SPC- A1 and A549 cells. q PCR and Western blot was used to detect the expression of epithelial- mesenchymal transformation( EMT) markers including epithelial cadherin( E-cadherin) and Vimentin. Results:SPRY4- IT1 was low- expression in NSCLC tissue and cells comparing to the normal tissue or cells( P 0. 01). Overexpression of SPRY4- IT1 reduced cells proliferation clongenic ability and invasiveness. SPRY4- IT1 significantly increased the E- cadherin expression level and decreased the Vimentin expression. Conclusion:long non- coding RNA SPRY4- IT1 low- expression may promote cancer cell proliferation and invasion via EMT pathway.
