雷公藤甲素对糖尿病肾病大鼠尿微量白蛋白排泄的影响 被引量:3

摘要 目的探讨中药雷公藤提取物雷公藤甲素(triptolide,TP)对糖尿病肾病(diabetic nephropathy,DN)大鼠尿微量白蛋白排泄的影响。方法 60只SD大鼠随机分为正常对照组(Control组,8只)和实验组(52只),实验组使用链脲佐菌素(streptozocin,STZ)建立DN模型(有4只大鼠血糖未达标,剔除实验)。将48只造模成功的大鼠按照随机数字表法分为4组,每组12只:DN组、TP低剂量组(Tl组,予TP 0.2mg·kg-1·d-1)、TP高剂量组(Th组,予TP 0.4mg·kg-1·d-1)、贝那普利治疗阳性对照组(Ben组,予贝那普利10mg·kg-1·d-1)。成模后第5周开始灌胃治疗,每天1次,共治疗4周。治疗前后检测大鼠的血糖和24h尿微量白蛋白。结果 DN组、Tl组、Th组、Ben组在糖尿病成模时血糖水平均较基础血糖明显升高(P<0.01),DN组、Tl组、Th组、Ben组各组间血糖水平比较差异均无统计学意义(均P>0.05)。成模4周时DN组、Tl组、Th组和Ben组24h尿微量白蛋白均较Control组增高(均P<0.01),提示DN已经发生;成模8周时Ben组、Tl组、Th组3组24h尿微量白蛋白均较DN组显著降低(P<0.01),Tl组和Th组比Ben组降低更明显(P<0.05)。结论 TP能显著减少DN大鼠尿微量白蛋白的排泄,是减少大鼠DN尿蛋白的有效药物。 Objective To investigate the effect of triptolide(TP)on urinary albumin excretion in rats with diabetic nephropathy(DN).Methods Sixty SD rats were randomly divided into negative control group(n=8)and experimental group(n=52).Rats in experimental group were given streptozocin to establish the model of DN(four rats were excluded from the study because of inadequate blood glucose levels).The 48 DN rats were randomly divided into four groups:DN group,0.2mg·kg-1·d-1 TP group(Tl group),0.4mg·kg-1·d-1 TP group(Th group),and 10mg·kg-1·d-1 benazepril group(Ben group),with 12 rats in each group.Intragastric administration was performed from week 5after successful modeling once daily for 4weeks.Detect blood glucose and 24-hour urinary in rats before and after the treatment.Results At model establishment,blood glucose levels increased in model rats compared with negative control group(P<0.01),but no significant differences were found among DN,Tl,Th and Ben groups(P>0.05).At 4weeks after model establishment,compared with negative control group,24-hour urinary albumin excretion increased in DN,Tl,Th and Ben groups(P<0.01).At 8weeks after model establishment,compared with DN group,24-hour urinary albumin excretion decreased in Tl,Th and Ben groups(P<0.01),and the decrease in Tl and Th groups was more obvious than that in Ben group(P<0.05).Conclusion TP can effectively reduce urinary albumin excretion in DN rats.
