不同产地黄芩ITS2和psbA-trnH条形码序列分析 被引量:5

Sequence Analysis of ITS2 and psb A-trnH DNA Barcode of Wild Populations of Scutellaria baicalensis Georgi from Different Producing Areas

摘要 为吉林省野生黄芩种质鉴定提供参考,为建立黄芩鉴定标准化体系提供依据。使用中药DNA条形码鉴定技术,对目的片段进行扩增并分析,比较不同产地黄芩样本ITS2和psbA-trnH条形码片段的差异,寻找鉴别位点。10个不同产地野生黄芩药材的ITS2和psbA-trnH序列比对结果显示,在232 bp(ITS2)+318 bp(psbA-trnH),共550 bp的序列比对中,仅检测到1个"G"位点的插入/缺失,该位点位于ITS2矩阵的第53 bp处。在吉林省长春、向海、前郭、镇赉、洮南5个地区的野生样品中检测到1个"G"位点的插入/缺失,其他5个省份的样品中无此位点。 The standardized and automated identification for Chinese medicinal materials is important for the safe use and international trade of medicinal plants,including the identification for genuine or non-genuine medicinal plants. Scutellaria baicalensis Georgi is a perennial herb of the family Lamiaceae. Root of the herb is one of the most important heat-clearing and dehumidifying drugs in traditional Chinese medicine. The wild populations of S. baicalensis are widely distributed from Northeast to Northwest China. The technology of DNA bar-coding was used to identify different germplasms of S. baicalensis. The ITS2 and psb A-trn H were selected as DNA barcode because of their quick evolutionary rate in the experiment. The genomic DNAs were extracted from wild population of S. baicalensis at ten districts,six provinces. The segments of two barcodes were amplified and aligned. The aligned sequences of ITS2 and psb A-trn H were 232 and 318 base pairs in length. The results showed that only one site of "G"insertion / deletion was found,which was located at the 53 th base in the ITS2 alignment. The site was detected in the samples of Xianghai,Qianguo,Zhenlai,Taonan,Changchun of Jilin province,but not in other samples from other five provinces. The site can be considered as reference to identify the wild S. baicalensis in Jilin province.
