怀地黄叶片离体再生及试管地黄诱导研究 被引量:1

摘要 目的通过筛选最适于怀地黄温85-5的叶片愈伤诱导、再生芽分化和试管地黄诱导的6-BA、NAA浓度,建立怀地黄离体再生和试管地黄诱导体系,为脱毒地黄苗和微型块根种子的培育奠定基础。方法分析在添加不同浓度NAA、6-BA情况下地黄叶片的愈伤诱导率、再生芽分化率、试管地黄诱导率和再生芽生根率。结果最适于地黄温85-5的叶片再生芽分化的激素浓度为NAA 0.1 mg/L、6-BA 2.0 mg/L。在NAA 0.1 mg/L、6-BA 2.0 mg/L时叶片愈伤的试管地黄诱导率最高,而且适于叶片和茎直接诱导试管地黄。添加0.1 mg/L的NAA有利于再生地黄植株壮苗和提高生根率。结论激素浓度对于地黄的愈伤诱导离体再生和试管地黄诱导均有显著的影响。 Objective To establish shoot regeneration and micro-tuberous induction system from leaves of Rehmannia glutinosa,analyzed the effects of hormone concentration on callus induction,shoot regeneration,and tuberous induction,etc.Provide a crucial foundation for cultivation of virus-free seedling and micro-tuberous of R.glutinosa.Methods Leaves of R.glutinosa 85-5 as explants,effects of different concentration of a-naphthalene acetic acid(NAA) and 6-benzylaminopurine(6-BA) on rates of callus induction,shoot regeneration,microtuberous induction and rooting were studied.Results It showed that the optimum concentration of hormone for shoot regeneration of Wen 85-5 was 0.1 mg/L NAA,2.0 mg/L 6-BA.The optimal concentration of hormone for micro-tuberous induction from leaf callus was 0.1 mg/L NAA and 2.0 mg/L 6-BA,it is also suitable for micro-tuberous induction from leaf and stem.Adding 0.1 mg/L NAA was beneficial to culturing sound seedlings and increase rooting rate.Conclusion The concentration of NAA and 6-BA were significant effects on shoot regeneration,rooting and tuberous induction from leaves of R.glutinosa.
