摘要
以42℃作为热应激模型温度、37℃作为对照温度,于42℃培养的卵巢细胞中分别添加富硒女贞子水煎液、齐墩果酸、蛋白质和多糖,采用四唑盐(MTT)法检测细胞增殖活性、流式细胞仪检测细胞凋亡、黄嘌呤氧化酶法检测细胞培养上清中超氧化物歧化酶(SOD)活性,探讨富硒女贞子及其主要成分对高温体外培养卵巢细胞生长、凋亡及抗氧化功能的影响。结果表明:富硒女贞子水煎液及多糖能显著提高热应激状态下卵巢细胞的增殖活性(P<0.05)、显著降低细胞的凋亡率(P<0.05)、显著提高细胞培养上清中SOD活性(P<0.05),且富硒女贞子效果明显优于女贞子;富硒女贞子蛋白质及齐墩果酸效果并不显著。说明富硒女贞子水煎液、多糖对热应激引起的卵巢细胞增殖活性降低及细胞凋亡有明显的改善作用,并能提高卵巢细胞的抗氧化功能。
In the experiment,the heat stress model temperature was set as 42℃ and the temperature 37℃ was set as the control,heat-stressed ovarian cells were cultured with different concentration of water extract,polysaccharides,protein and oleanolic acid extract of Se-LLF.The growth and apoptosis rate of cultured ovarian cells were respectively detected by MTT and flow cytometry,and the content of SOD in the supernatant fluid of cultured ovarian cells was detected by xanthine oxidase to investigate the effects of se-enriched Ligustri Lucidi Fructus on proliferation,apoptosis and antioxidant activity of ovarian cells cultured under heat circumstance in vitro.The results showed that the proliferation of ovarian cells cultured under high temperature was significantly promoted in each experimental concentration of water extract and polysaccharides extract of Se-LLF(P〈0.05).The apoptosis rate decreased significantly(P〈0.05),the activity of SOD was increased significantly(P〈0.05),and the results were better than those of the ordinary Ligustrum Lucidum group.In conclusion,water and polysaccharides extract of Se-LLF can not only improve the proliferation and antioxidant capacity of heat-stressed ovarian cells,but also can inhibit their apoptosis.
