摘要
目的:探讨壮骨止痛方抗骨质疏松作用机制。方法:雌性SD大鼠40只随机分为模型组、壮骨止痛方组、阳性对照组、假手术组,给药5 d后制备含药血清。获取新生SD乳鼠成骨细胞,培养至第3代,分别加入4组血清。培养6 d后,测定碱性磷酸酶(ALP)活性,用免疫组化法检测各组细胞中cyclin D1蛋白表达;18 d后进行茜素红染色,观察各组钙化结节。结果:壮骨止痛方组ALP活性以及cyclin D1蛋白表达强于阳性对照组,表明成骨细胞增殖旺盛。结论:壮骨止痛方可提高体外培养成骨细胞cyclin D1表达,对骨质疏松症有治疗效果。
Objective:To study the mechanism of anti-osteoporosis effect of Zhuanggu Zhitong fomula. Methods:40 female SD rats were randomly divided into model group,positive control group,sam operation group and Zhuanggu Zhitong fomula group. Containing serum was prepared 5 d after administration. Osteoblasts of SD neonatal rats were obtained and cultured to the third generation,then containing serum of above groups were respectively added in. 6 d after culture,alkaline phosphatase activity and protein expression of cyclin D1 were detected. 18 d after culture,calcified nodules were observed through alizarin red staining. Result:Compared with positive control group,ALP activity and protein expression of cyclin D1 of Zhuanggu Zhitong fomula group were increased(P〈0.05). Conclusion:Zhuanggu Zhitong fomula can improve osteoblast cyclin D1 expression and has therapeutic effect on osteoporosis.
