慢性非细菌性前列腺炎小鼠前列腺中α_1-受体亚型的表达变化 被引量:3

Expression of α_1-adrenoceptor subtypes in the mouse with chronic abacterial prostatitis

摘要 目的:复制小鼠慢性非细菌性前列腺炎(CAP)模型,研究前列腺炎发展过程中α1肾上素能受体(α1-AR)亚型的表达及其与前列腺中炎症介质TNF-α的关系。方法:使用大鼠前列腺蛋白提纯液辅以双重免疫佐剂,造成实验性慢性非细菌性前列腺炎(CAP)小鼠模型,在造模后45天解剖小鼠,获得前列腺组织样本,抽提RNA转录为cDNA,运用实时荧光逆转录聚合酶链反应检测组织标本中α1a-AR、α1b-AR、α1d-AR、TNF-α的mRNA的表达,观察CAP小鼠α1-AR三种亚型的表达变化,并观察其与前列腺中炎症介质TNF-α的关系。背侧组织做病理切片观察,进一步判别前列腺的炎症程度。结果:造模小鼠均出现排尿障碍等症状;病理切片观察到炎症细胞浸润;造模小鼠中TNF-α明显升高,但α1a-AR、α1b-AR、α1d-AR无明显变化。结论:慢性前列腺炎小鼠模型复制成功,CAP的炎症程度与TNF-α的表达成正相关关系,与α1a-AR、α1b-AR、α1d-AR无明显关系。 Objective:To make a kind of chronic abacterial prostatitis(CAP)mice,and try to find the relationship between α1-adrenoceptor( α1-AR)subtypes and prostate inflammatory mediators TNF-α.Method:We use prostate protein purification solution from SD rat to cause a kind of experimental CAP mouse model with two immune adjuvants.We killed mice after 45 days to get the prostatits tissue samples.And then,RNA was extracted and transcribed into cDNA.Later,we used real time reaction to detect the expression of α1a-AR, α1bAR, α1d-AR,TNF-α,and found the expression changes in three α1-AR subtypes as well as the relationship with TNF-α.We also did some pathological observation to define the degree of inflammation in prostate.Result:The symptoms such as building urination disorders occurring in CAP mice and pathological inflammatory cell infiltration were observed.In realtime result,TNF-αhad a higher expression in CAP group,while α1a-AR, α1b-AR and α1d-AR didnt show the significance.Conclusion:We have copied a kind of CAP mice.The degree of inflammation is positively related to the expression of TNF-α,but not associated with α1a-AR, α1b-AR or α1d-AR.
