针刺对卒中后抑郁大鼠海马超微结构的影响 被引量:6

Influence of acupuncture on hippocampus ultrastructure of rats with post-stroke depression

摘要 目的观察针刺对卒中后抑郁(PSD)大鼠海马超微结构的影响,以期从形态学角度探讨其作用机制。方法将120只SD大鼠随机分为空白组、模型组、药物组、针刺组,其中空白组20只,模型组30只,药物组和针刺组各35只,采用大脑中动脉线栓阻塞法联合慢性不可预知性温和应激处理方法制备PSD模型。药物组予氟西汀2 mg/kg溶入生理盐水灌胃,针刺组选取百会、风府、神门、太冲穴针刺,留针20 min,期间行针1次,以上治疗每日1次,7 d为1个疗程,共3个疗程,每个疗程之间休息1 d。在光镜下观察大鼠海马病理变化,并在透射电镜下观察大鼠海马超微结构的变化。结果模型组大鼠海马神经元超微结构发生明显改变,神经元损害严重;针刺组和药物组大鼠海马神经元超微结构变化相对较小,神经元受损程度明显轻于模型组。结论针刺可以逆转PSD所造成的大鼠海马神经元的损伤,这可能是针刺抗抑郁的作用机制之一。 Objective To observe the effect of acupuncture on hippocampus ultrastructure of rats with poststroke depression( PSD) and explore the mechanism of action from the morphological. Methods 120 SD rats were randomized into blank group( 20 cases),model group( 30 cases),drugs group( 35 cases) and acupuncture group( 35 cases). PSD models were made by occlusion of middle cerebral artery( MCAO) and chronic unpredictable mild stress( CUMS) processing. The drugs group was given gavage administration with 2 mg / kg fluoxetine. Points of ‘Baihui’( DU 20),‘Fengfu’( DU16),‘Shenmen’( HT7),‘Taichong’( LR3) were chosen to acupuncture,needle retaining for 20 minutes and manipulating the needle one time during retaining. The treatment was taken once every day,7 days as a course and prolonged for 3 courses,1 day off between courses. Hippocampus pathological change was observed under light microscope and hippocampus ultrastructure change was observed under transmission electron microscope. Results Hippocampus ultrastructure in model group changed greatly with se-rious neuron injury,while that in acupuncture group and drug group changed less with mild injury. Conclusion Acupuncture can improve the damage on neuron,it may be the mechanism of anti-depression.
